    Abstract: The invention provides a method using a hemocyte preparation, for example, Limulus amebocyte lysate, for detecting in a single assay the presence of at least one of a Gram negative bacterium, a Gram positive bacterium, and a fungus in a sample of interest. The method exploits the differential reactivity of Gram negative bacteria, Gram positive bacteria, and fungi with the hemocyte preparation to produce measurable changes in a property, for example, an optical property, of the mixture. Because the Gram negative bacteria, Gram positive bacteria and fungi each produce different changes in a given property, for example, an optical property, it is possible to classify the type of microorganism present in the sample of interest.

The cartridge comprises (i) a housing defining a sample inlet, an optical cell, an outlet, a first conduit in fluidic communication with the sample inlet and the optical cell, and a second conduit in fluidic communication with the outlet and the optical cell, (ii) an addressable array disposed within the optical cell; and (iii) a reagent dried upon a fluid contacting surface of at least one of the sample inlet and the first conduit, such that, when a fluid sample is applied to the fluid inlet, the fluid sample mobilizes and transports the reagent to the optical cell. The invention also provides a method of detecting one or more analytes in a fluid sample of interest.
