    Abstract: The invention provides methods and compositions for the detection and/or quantification of a microbial contaminant, for example, a bacterial endotoxin or a glucan, in a sample. In particular, the invention provides a test cartridge useful in the practice of hemocyte lysate-based assays for the detection and/or quantification of a microbial contaminant in a sample. In addition, the invention provides methods of making and using such cartridges. In addition, the invention provides a rapid, sensitive, multi-step kinetic hemocyte lysate-based assay for the detection and/or quantification of a microbial contaminant in a sample. In addition, the invention provides a glucan-specific lysate that can be used in a variety of assay formats, including, for example, a test cartridge, optionally configured to perform a kinetic assay.

    Abstract: This invention is directed to pharmaceutical and cosmetic compositions comprising anti-lipopolysaccharide (anti-LPS) factor proteins derived from horseshoe crabs, either in the native form or produced by recombinant means. The pharmaceutical formulations, which may include anti-LPS factor proteins alone or in combination with other antimicrobials, may be used in the treatment of gram-negative bacterial infections, endotoxemia, septic shock, gram-positive bacterial infections, and yeast infections. The anti-LPS factor protein-containing pharmaceuticals can be formulated for systemic or topical administration. They may also be used to control mold growth. Anti-LPS factor proteins can be used in cosmetic compositions or skin or hair preparations as antimicrobial preservatives, either alone or in combination with conventional preservatives, to prevent or control the growth of bacteria, yeast and mold.

    Abstract: The invention is directed to a process and a medium for simultaneous determination of the number and presence of living fecal coliform and Escherichia coli in a sample comprising 6-O-alpha-D-galactopyranosyl-D-glucose or isopropyl-beta-D-thiogalactoside as a galactosidase inducer and methyl-beta-D-glucuronide as a glucuronidase inducer. The sterile semi-solid medium also comprises non-target bacterial inhibitors, target bacterial enhancers, and multiple fluorogen and/or chromogan substrates that produce color and fluorescence upon cleavage by a specific enzyme expressed by the target bacteria in which expression is enhanced. The simultaneous detection of total coliforms via its expression of beta-galactosidase, and Escherichia coli as the target bacteria via its expression of beta-galactosidase and beta-glucuronidase is achieved rapidly and efficiently using this medium.
