    Abstract: Embodiments of the disclosure provide systems and methods for positioning a vehicle. The system includes a communication interface configured to receive point cloud frames with respect to a scene and initial pose data of a vehicle captured by sensors equipped on the vehicle as the vehicle moves along a trajectory. The system also includes a storage configured to store the point cloud frames and the initial pose data. The system further includes a processor configured to estimate pose information of the vehicle associated with each of the point cloud frames based on the initial pose data and the point cloud frames. The processor is also configured to adjust the estimated pose information of the vehicle based on a model. The model includes a spatial relationship and a temporal relationship among the plurality of point cloud frames. The processor is further configured to position the vehicle based on the adjusted pose information.

    Abstract: The subject invention concerns materials and methods for culture of cell aggregates. The subject invention utilizes three-dimensional (3-D) inserts comprising micro-channels having selected dimensions. The inserts are provided in or on tissue culture plates that can be supported on a programmable rocking platform/station, thereby providing for a hydrodynamic environment that promotes 3-D aggregation of cells cultured on the plates. In one embodiment, one or more micro-channels are provided in a 3-D insert that can be provided in or placed within the tissue culture plate. Cells are placed in the micro-channels of the inserts in the tissue culture plates that comprise a suitable cell growth medium for the particular type of cell. The supporting rocker is programmed to provide motion that generates hydrodynamic conditions that support 3-D cell aggregation and long-term culture. Aggregated cells can be harvested and isolated from the micro-channels of the 3-D inserts in the tissue culture plate at suitable times.

A laser is then used focused onto the grating to determine the strain on the surface by determining the variation of the grating wavelength due to the strain on the surface. The strain information is essentially carried by the grating, in terms of grating wavelength, because it varies according to the volume change of the underlying substrates. By scanning the surface grating with the small laser size, a high resolution strain map of the surface can be produced. The induced strain is related to the grating wavelength variation, which leads to the diffraction angle variation that is captured by the strain sensing measurements.

    Abstract: The subject invention pertains to acellular extracellular matrix (ECM) and methods for preparing it. In one embodiment, the ECM is prepared from 3-D aggregates of pluripotent stem cells or from embryoid bodies. In one embodiment, the ECM is in the form of acellular microspheres. The stem cells used in the present invention can be embryonic stem cells (ESC). In a specific embodiment, the stem cells or embryoid bodies are from human. The ECM of the invention can be mass produced using PSC aggregates or embryoid bodies in large scale bioreactors. The acellular ECM produced using the present invention can be used for injection into a person or animal to regenerate tissue and/or treat a disease or condition. A method of the invention comprises growing stem cells in suspension culture as aggregates, e.g., in a bioreactor. Acellular ECM is then prepared from the cell aggregates by decellularization.

The subject invention integrates a cell aggregation process in a scalable bioreactor system. In one embodiment of the method, thermally responsive microcarriers (TRMs) are utilized in conjunction with a bioreactor system. Cells are cultured in a container or vessel in the presence of the TRMs wherein cells adhere to the surface of the TRMs. Once cells are adhered to the TRMs they can be cultured at a suitable temperature for cell growth and expansion, e.g., at about 37° C.
