Polypeptide derivatives of human granulocyte colony stimulating factor

Novel hG-CSF polypeptide derivatives having an amino acid sequence derived from the amino acid sequence of the human granulocyte colony stimulating factor polypeptide by substitution of at least one amino acid by a different amino acid and/or deletion of at least one amino acid, recombinant plasmids containing a DNA fragment insert coding for any of these hG-CSF polypeptide derivatives, microorganisms carrying one of such plasmids, methods of producing the hG-CSF polypeptide derivatives using the microorganisms, a monoclonal antibody binding to the hG-CSF polypeptide derivative, and chemically modified hG-CSF or derivatives thereof are disclosed.

Claims

1. A modified polypeptide having human granulocyte colony stimulating factor (hG-CSF) activity comprising a polypeptide having an amino acid sequence of the hG-CSF polypeptide wherein the amino acids sequence is derived from the amino acid sequence of the hG-CSF, which is defined in the Sequence listing by SEQ ID: No. 1, by substitution of at least one amino acid out of the first to sixth amino acids on the N terminus side and optionally the seventeenth amino acid thereof with proviso that a polypeptide wherein the amino acids in the first, third, fourth, fifth and seventeenth position from the N terminus side are Ala, Thr, Tyr, Arg and Ser, respectively, is excluded, and with at least one amino group thereof modified by a group of the formula ##STR38## wherein R.sub.1 is an alkyl or alkanoyl group; n is an optionally variable positive integer;

X is O, NH or S; ##STR39## where R.sub.3 is OH, Cl or O--(CH.sub.2 CH.sub.2 O).sub.n --R.sub.1, where R.sub.1 and n are as defined above, Y may not be present or represents Z--(CH.sub.2).sub.p CO, where Z is O, S or NH and p is an optionally variable positive integer;
or R.sub.2 is (CO).sub.m --(CH.sub.2).sub.1 CO, where m is 0 or 1 and 1 is an optionally variable positive integer.

2. The modified polypeptide of claim 1, wherein R.sub.1 is a C.sub.1-18 alkyl or alkanoyl group.

3. The modified polypeptide of claim 1, wherein n is a positive integer of not more than 500.

4. The modified polypeptide of claim 1, wherein l is a positive integer of not more than 100.

5. The modified polypeptide of claim 1, wherein the group of formula (I) has a molecular weight of not more than 30,000.

6. A lyophilized preparation containing the modified polypeptide of claim 1.

7. A leukocyte growth promoting agent comprising the modified polypeptide of claim 1.

8. A modified polypeptide derived from the hG-CSF human granulocyte colony stimulating factor (hG-CSF) polypeptide of SEQ ID NO: 1 polypeptide by deletion, wherein a peptide comprising the first to fourth, first to fifth, first to sixth, first to seventh or first to eleventh amino acids on the N terminus position of said hG-CSF polypeptide is missing, and optionally in addition to amino acid deletion, the seventeenth amino acid cysteine of said hG-CSF polypeptide has been replaced by serine, and with at least one amino group thereof modified by a group of the formula ##STR40## wherein R.sub.1 is an alkyl or akianoyl group; n is an optionally variable positive integer;

X is O, NH or S; ##STR41## wherein R.sub.3 is OH, Cl or O--(CH.sub.2 CH.sub.2 O).sub.n --R.sub.1, where R.sub.1 and n are as defined above, Y may not be present or represents Z--CH.sub.2).sub.p CO, where Z is O, S or NH and p is an optionally variable positive integer;
or R.sub.2 is (CO).sub.m --(CH.sub.2).sub.1 CO, where m is 0 or 1 and 1 is an optionally variable positive integer.
