Abstract: Method for controlling a liquid chromatography system comprising a system pump and a column in fluid communication with the system pump by a fluid flow path, the method comprising the steps: registering the system pressure at a flow path position close to the system pump, controlling the operation of the system pump in response to the registered system pressure, estimating a pre-column pressure based on the registered system pressure, the characteristics of the flow path, and the viscosity and flow-rate of the liquid in the system, and controlling the operation of the system pump in response to the estimated pre-column pressure.

Abstract: The present invention relates to a method of preparation of formulations of gadolinium metal complexes of the macrocyclic chelator DOTA, which further comprise a small excess of free DOTA. The method uses controlled conditions such that excess gadolinium is present as a precipitate of gadolinium oxide, with filtration to remove the excess, prior to the addition of a defined excess of DOTA chelator. Also provided is a method of preparation of MRI contrast agents based on Gd-DOTA.

Abstract: Disclosed herein is provided a virus purification and formulation process for purifying a flavivirus represented by one of a Yellow Fever Virus, Japanese Encephalitis virus, Dengue virus, and West Nile virus. The highly purified flavivirus virus product is characterized as having a low level of sucrose without significant virus loss such as that which is typically encountered by prior art virus purification processes. The disclosed process captures and purifies the virus, separating it from the host cell proteins and DNA, and leaving the host cell proteins and DNA behind. The process also can be used to inactivate and/or concentrate the virus sufficiently for use in formulations.
