Abstract: A microbial oil comprising: a specific amount of at least one polyunsaturated fatty acid having at least 20 carbon atoms in fatty acid alkyl ester form and/or in free fatty acid form; and specific amount of thermally-produced fatty acid having from 16 to 22 carbon atoms in a fatty acid alkyl ester form and/or a free fatty acid form. A production method thereof comprising: providing a starting oil containing at least one polyunsaturated fatty acid having at least 20 carbon atoms in an alkyl ester form and/or a free fatty acid form obtained from microbial biomass; performing a rectification of the starting oil under specific conditions; and obtaining the aforementioned microbial oil. A concentrated microbial oil obtained using the production method described above, and a production method thereof. An agent for treating or preventing an inflammatory disease comprising the microbial oil or the concentrated microbial oil.
Abstract: The present invention provides a method of producing a composition containing docosahexaenoic acid as a constituent fatty acid of glycerides, comprising hydrolyzing a feedstock oil containing glycerides comprising docosahexaenoic acid as a constituent fatty acid by action of a first lipase and a second lipase, thereby increasing the proportion of docosahexaenoic acid in glyceride fractions, wherein the first lipase is at least one lipase selected from the group consisting of a lipase derived from a microorganism of the genus Thermomyces, a lipase derived from a microorganism of the genus Pseudomonas, a lipase derived from a microorganism of the genus Burkholderia, and a lipase derived from a microorganism of the genus Alcaligenes, and wherein the second lipase is a partial glyceride lipase.

Abstract: A method for producing a microbial oil includes steps of: genetically modifying a labyrinthulid by disrupting and/or silencing a gene, or by transforming another gene in addition to the disruption and/or gene silencing of the gene, and culturing the labyrinthulid, such that a fatty acid composition accumulated in the labyrinthulid comprises an increased EPA content; and collecting the microbial oil having the increased EPA content from the labyrinthulid. The labyrinthulid before the modification is selected from (A) a labyrinthulid belonging to the genus Parietichytrium or genus Schizochytrium and having very weak or no activity of producing PUFAs via a PUFA-PKS pathway; and (B) a labyrinthulid belonging to the genus Thraustochytrium in which a host PUFA-PKS gene is disrupted or silenced to a very weak level. The increased EPA content is preferably not less than 11.5% of a total fatty acid composition.

It is preferable for the DGLA to be present as a glyceride, a phospholipid, or an alkyl ester. The amount of DGLA contained in the composition for external use is 0.1-50 wt %.
