Abstract: The invention provides compositions and methods for high-efficiency genome editing. In some aspects, the invention provides retron-guide RNA cassettes and vectors comprising the cassettes. Also provided are host cells that have been transformed with the vectors. In other aspects, the invention provides retron donor DNA-guide molecules. In some other aspects, methods for genome editing and the screening of genetic loci are provided. In further aspects, methods and compositions are provided for the prevention or treatment of genetic diseases. Kits for genome editing and screening are also provided.

Abstract: A device (“the ExoTIC device”) for the isolation of extracellular vesicles from an extracellular vesicle-containing sample in which the sample is flowed through a membrane in a flow chamber to capture and purify the extracellular vesicles on the membrane. Abstract: A method and device for extracting information from acoustic signals receives acoustic signals by a microphone, processes them in an analog front-end circuit, converts the processed signals from the analog front-end circuit to digital signals by sampling at a rate of less than 1 kHz or more preferably less than 500 kHz; and processes the digital signals by a digital back-end classifier circuit. Abstract: The present methods are exemplified by a process in which maternal blood containing fetal DNA is diluted to a nominal value of approximately 0.5 genome equivalent of DNA per reaction sample. Digital analysis is then be used to detect aneuploidy, such as the trisomy that causes Down Syndrome. Since aneuploidies do not present a mutational change in sequence, and are merely a change in the number of chromosomes, it has not been possible to detect them in a fetus without resorting to invasive techniques such as amniocentesis or chorionic villi sampling. Digital amplification allows the detection of aneuploidy using massively parallel amplification and detection methods, examining, e.g., 10,000 genome equivalents.

Abstract: Systems and methods for spatial graph convolutions in accordance with embodiments of the invention are illustrated. Abstract: Turning now to the drawings, systems and methods to enhance RNA transcription in a cost-effective manner and uses thereof are provided. One of the most common enzymes for RNA transcription is T7 RNA polymerase. Many embodiments increase RNA yield in transcription reactions by adding ribonucleoside vanadyl complex (VRC) to the transcription reaction. Various embodiments use VRC at low concentrations in an RNA transcription reaction. Reactions in accordance with many embodiments are capable of increasing RNA yield by approximately 2-fold or more.

Abstract: A method for radiation therapy treatment planning includes: a) defining a decision variable search space by projecting an initial seed point onto a pareto front, where the pareto front and decision variable search space are defined in the same coordinate space; b) projecting search points in the decision variable search space to the pareto front using a one-dimensional search algorithm to produce projected points on the pareto front; c) updating the search points by performing a gradient-free optimization that evaluates a treatment planning scoring function at the projected points on the pareto front; and d) repeating steps (b), (c) to search within the decision variable search space until a convergence criterion is satisfied, thereby producing a pareto optimal and clinically acceptable treatment plan.
