    Abstract: The present disclosure describes two click reactions to sequentially polymerize and then functionalize hydrogel microparticles to provide therapeutic agent-functionalized crosslinked hydrogel microparticles. The two click reactions have one common reactive group, and are non-interfering with respect to one another. The therapeutic agents can be released from the hydrogel microparticles over time as the hydrogel degrades, or the therapeutic agents can be released from the hydrogel microparticles by enzymatic degradation or hydrolysis. In some embodiments, the biological function of the therapeutic agents can be preserved or enhanced when covalently bound to the hydrogel microparticles. The present disclosure also provides methods for the production of hydrogel cell culture substrates.     Abstract: A non-contact photoacoustic spectrophotometry system is configured to measure an absorption spectrum of a material. The system includes a modulated light source such as tunable pulsed laser that generates laser pulses to produce photoacoustic signals in the material. A non-contact detector monitors the surface of the container for the material. The detector includes a second light source, such as a continuous wave laser, focused on the surface of the container, and transmits reflected light to an interferometer, for example, a Sagnac interferometer. The interferometer produces an interference signal from the received light that is proportional to the acoustic pressure, which is transmitted to a computer to calculate an absorption coefficient. Using a plurality of wavelengths from the tunable pulsed laser, an absorption spectrum may be generated.

    Abstract: A pulsed beam of NIR excitation light is projected into a sample (345) at an oblique angle and scanned by a scanning element through a volume in the sample. 2-photon excitation excites fluorescence within the sample. The fluorescence is imaged onto an intermediate image plane that remains stationary regardless of the orientation of the scanning element. The image is captured by a linear array of light detecting elements (392) or a linear portion of a rectangular array. At any given position of the scanning element, the linear array (or portion) images all depths simultaneously. A plurality of images are captured for each of a plurality of different orientations of the scanning element. The orientation of the scanning element is controlled to move in a two dimensional pattern, which causes the beam of excitation light to sweep out a three dimensional volume within the sample.
