    Abstract: The present invention relates to the method of measuring a biological ligand (1) in a sample containing an amylase (1) derived from a higher animal. This method is characterized by employing a combination of an amylase (2) not contained in the sample and an antibody against the ligand (1) or a ligand (2) capable of reacting with the antibody and an amylase inhibitor of which inhibitory activity against the amylase (1) is stronger than that against the amylase (2). The method of the invention utilizes the emphasis of steric hindrance of the amylase (2) reaction which is carried out at the boundary between solid phase and liquid phase and the specific removal of the influence of the amylase (1) by using the above amylase inhibitor. This method is suitable as a clinical test for the determination of physiological substances and trace components in a humoral fluid such as serum.

    Abstract: Disclosed is a method for detection for serum antibody and/or microbial surface antigen by radial partition immunoassay. The method of this invention is applicable to (a) the evaluation of a clinical specimen for identification of a microorganism; (b) antimicrobial sensitivity assays for determination of an efficacious antibiotic for use against a specific microorganism; (c) a semi-quantitative determination of viral surface antigen; and, (d) a quantitative method for the determination of the presence of serum antibodies to microbial antigens. In each of the foregoing applications, the analyte of interest can be immobilized within a porous matrix (solid phase) by simple pipetting of the sample onto the prepared matrix. Appropriate reagents are subsequently applied to the matrix to effect immunochemical interaction of a labeled binding material to the surface antigen (or antibody) of the analyte of interest. The portion of the matrix within which such interaction takes place is termed the "reaction zone".

    Abstract: A method for assaying for a ligand analyte which is a member of a specific binding pair ("sbp member") consisting of ligand and its complementary receptor is disclosed. The ligand analyte has a binding site in common with an interfering substance present in a sample suspected of containing the analyte. The interfering substance has at least two binding sites. The method comprises combining in an assay medium without intervening separation (1) the sample, (2) a blocking receptor which does not bind to the ligand and does bind to the interfering substance, thereby blocking the binding of a common receptor to the interfering substance, and (3) a common receptor which binds to the common binding site. Any additional members of a signal producing system capable of producing a detectable signal in relation to the amount of analyte in the sample are added to the assay medium. Next, the assay medium is examined for the presence of a detectable signal.

The labels of this invention have greatly improved stability in aqueous solutions and do not prematurely agglomerate during storage. The antibody (conjugated and/or immobilized) comprises multiple monoclonal antibodies from different cell lines so that the specificity of the assay is enhanced, and the possibility of unrecognized antigen fragments is reduced.
