  • Publication number: 20240425559
Abstract: Recombinant TGF-?2 monomers engineered to prevent dimerization and block TGF-? signaling are described. The engineered monomers lack the ability to bind and recruit TGF-? type I receptor (TbRI), but retain the capacity to bind the high affinity TGF-? type II receptor (TbRII). The TGF-?2 monomers also include additional modifications that increase their affinity for TbRII, reduce their aggregation and/or improve their folding. Nucleic acid molecules and vectors encoding the recombinant TGF-?2 monomers are also described. Isolated cells, such as T cells, can be re-programmed with a TGF-?2 monomer-encoding nucleic acid or vector to secrete the monomer. Use of the recombinant TGF-?2 monomers and/or cells producing the recombinant TGF-?2 monomers, to inhibit TGF-? signaling, such as to treat disorders associated with aberrant TGF-? signaling, are also described.

  • Publication number: 20240430555
Abstract: A pipe inspection system includes a cable storage drum and a housing configured to removably receive and rotatably support the cable storage drum. A push-cable with a plurality of conductors is stored in the drum. An inspection camera may be attached to a distal end of the push-cable. The drum includes a hollow core outer cone configured to accept an inner core removable module. The removable module may include electrical, mechanical, wireless, satellite, and power/battery interfaces. The module mates with a contact plate sealed inside the inner cone. The inner core components, including the battery module, are stationary relative to the inner core. The inner core is free to rotate bidirectionally inside the outer cone as the camera is being fed into, and out of a pipe or conduit for inspection. Slip-rings are not required to maintain connectivity because the electronics and battery module are stationary relative to each other.

  • Publication number: 20240427672
Abstract: A method for execution on a storage network begins with receipt of a data object for storage by a plurality of users and continues with a storage network processing unit determining whether the data object is associated with a replica storage node of a plurality of replica storage nodes associated with the storage network. When the data object is associated with a replica storage node, the method continues by facilitating storing the data object in each replica storage node of the plurality of replica storage nodes and receiving a plurality of access responses from at least some replica storage nodes of the plurality of replica storage nodes. The method then continues by determining whether an access response threshold number of access responses have been received and finally, in response to a determination that an access response threshold number of access responses have been received, transmitting an acknowledgement to the plurality of users.
