    Abstract: The present invention relates to a method and system for determining Copy Number Variations (CNVs) in a genomic test sample including target amplicons and a reference amplicons. Specifically, nucleic acid melting curves are generated for the test sample. A mathematical model is fitted to each of the nucleic acid melting curves to separate target and reference melting reactions within the measured nucleic acid melting curve. The fitting parameters of the mathematical model are calculated. A CNV of the test sample is determined based on the fitting parameters of the mathematical model corresponding to the target and reference melting reactions.

In one method, image data is gathered using 3D image sensors, while illumination of an area of interest is alternated with projection of a pattern onto the area of interest. The captured image data is used to display a 3D true color image; a monochrome grid on surfaces of the area of interest; a 3D true color image with a monochrome grid on surfaces of the area of interest; a 2D or 3D false color image where different colors identify different depths of regions of the area of interest; and/or a 2D or 3D true color image of an area of interest with features in the area of interest located a first depth relative to the image sensors highlighted with a common color. In another method, 3D image data is used to generate a 2D image that highlights features at a common depth using the same color.

    Abstract: Closed-loop melt pool size control for additive manufacturing is integrated with site-specific changes to a controller set-point. Site-specific control enables localized control of bead geometry and material properties in an additive manufacturing system, thus offering enhanced defect mitigation capabilities when compared with constant setpoint technologies. Trigger points, generated by the projection of a secondary geometry onto a primary geometry, mark locations of the volume of a part under manufacture where site-specific changes in setpoint occur. Through this technique, it is possible to manufacture a specific geometry that occurs beyond a predefined toolpath of a print head. Implications of this capability extend beyond localized control of bead geometry to potential mitigations of defects and functional grading of component properties.

The monitor and control system receives various sensor data from the operation of the pump, including the pump crank position, and executes a pump control equation or model based on the pump sensor data, pump load data and/or pump speed data. Pump control equations or models are specific to the design and dynamic operation of the pump, incorporating the number of plungers, pump dynamics, motor lag and motor dynamics, etc. Using the pump control equations or models, the monitor and control system determines control commands for the pump motor to reduce or eliminate the oscillatory discharge pressure at the pump.
