    Abstract: The present invention relates to the field of biotechnology, in particular to a DNA polymerase mutant and the use thereof. Compared with a wild type, the mutant provided in the present invention has a significantly improved DNA polymerization activity, significantly improved affinity to DNA and amplification uniformity; and has good effects in the amplification of multiplex PCR, high-GC templates and complex templates. A hot-start-version DNA polymerase is further prepared by means of using antibodies or chemical modifications, wherein the yield of the DNA polymerase is significantly improved, while the primer dimer is significantly decreased. The prepared mutant hot-start DNA polymerase can be applied to PCR amplification of multiplex PCR, high-GC templates and complex templates.

    Abstract: A control device includes a first capacitor, a drive controller, a constant current source, a discharge-controlled current source, a current mirror, and a sample-and-hold circuit. The constant current source generates a constant current. The discharge-controlled current source is coupled to the constant current source and the first capacitor. The current mirror is coupled to the first capacitor and the primary side of a primary-side switch. The current mirror generates a copy current, thereby generating a copy voltage across the first capacitor. When the drive controller turns on the primary-side switch, the sample-and-hold circuit drives the discharge-controlled current source to sample and hold a control current from the constant current and the copy current. When the node voltage is higher than the copy voltage, the comparison circuit drives the drive controller to turn off the primary-side switch.

    Abstract: An etch bias direction is determined based on a curvature of a contour in a substrate pattern. The etch bias direction is configured to be used to enhance an accuracy of a semiconductor patterning process relative to prior patterning processes. In some embodiments, a representation of the substrate pattern is received, which includes the contour in the substrate pattern. The curvature of the contour of the substrate pattern is determined, and an etch bias direction is determined based on the curvature by considering curvatures of adjacent contour portions. A simulation model is used to determine an etch effect based on the etch bias direction for an etching process on the substrate pattern.

    Abstract: A method for chemical-biological cascade synthesis of L-phosphinothricin is carried out as follows: 3-(methylethoxyphosphinyl) ethyl propionate is synthesized by addition reaction from diethoxymethylphosphine and acrylic acid, then a condensation reaction is carried out with 3-(methylethoxyphosphinyl) ethyl propionate and sodium ethoxide as reactants, then the product is subjected to a hydrolysis reaction with diethyl oxalate to synthesize 4-(hydroxymethylphosphinyl)-2-oxobutyric acid, and finally, L-phosphinothricin is catalytically synthesized by taking 4-(hydroxymethylphosphinyl)-2-oxobutyric acid as a raw material, and using highly active and stable wet cells co-expressing phsophinothricin dehydrogenase and alcohol dehydrogenase or co-expressing a phsophinothricin dehydrogenase mutant and alcohol dehydrogenase as a biocatalyst, thereby solving the problems of existing L-phosphinothricin synthesis being tedious, low asymmetric amination reduction activity and poor stability.
