  • Publication number: 20240026393
Abstract: Muconic acid is a molecule that can be converted into direct replacement chemicals for incumbent petrochemicals and performance-advantaged bioproducts. Disclosed herein are Pseudomonas putida KT2440 that are engineered to convert glucose and xylose, the primary carbohydrates in lignocellulosic hydrolysates, to muconic acid using a model-guided strategy to maximize the theoretical yield. Using adaptive laboratory evolution (ALE) and metabolic engineering in a strain engineered to express the D-xylose isomerase pathway, we demonstrated that mutations in the heterologous D-xylose:H+ symporter (XylE), increased expression of a major facilitator superfamily transporter (PP_2569), and overexpression of aroB encoding the native 3-dehydroquinate synthase, enable efficient muconic acid production from glucose and xylose simultaneously. Using the rationally engineered strain, we produced 33.7 g/L muconate at 0.18 g/L/h and a 46% molar yield (92% of the maximum theoretical yield).

  • Publication number: 20240029547
Abstract: A method includes accessing a first dataset including aerial imagery data and infrared data, accessing a second dataset including property boundary data, and identifying property boundaries associated with a geographic area. A plurality of models is applied to the datasets to identify and compute information of interest. Based on the first dataset and constrained by the property boundaries, a building detection model can be applied to identify a building, and a vegetation detection model can be applied to identify one or more areas of vegetation. An estimated distance can be determined between each of the areas of vegetation and the building as separation data, which can be compared to a defensible space guideline to determine a defensible space adherence score. A wildfire risk map can be generated, including the defensible space adherence score associated with the geographic area.

  • Publication number: 20230326022
Abstract: A method includes receiving an input histology image, processing, using a cell classification model, the input histology image to generate one or more lymphocyte density maps within the input histology image, and performing morphological image processing on the one or more lymphocyte density maps to identify one or more TLS regions within the input histology image. Each TLS region is represented by a respective cluster of lymphocyte cells. For each corresponding TLS region of the one or more TLS regions identified in the input histology image, the method also includes extracting, from the respective cluster of lymphocyte cells, a respective set of TLS features, and processing, using a TLS classification model, the respective set of TLS features to classify the corresponding TLS region as one of a first TLS maturation state, a second TLS maturation state, or a third TLS maturation state.

  • Publication number: 20220213662
Abstract: A dynamically deployable, low visibility pneumatic cofferdam for flood mitigation is disclosed. The cofferdam includes a first planar component, a second planar component, an inflatable hermetic airbag sandwiched therebetween, and connection members attached to the first planar component, a second planar component to form a cofferdam unit. The connection members are adjustable to apply and maintain a desired amount of pressure on the airbag to enable a watertight seal to be formed. The cofferdam units may be sized and designed to sealingly mate adjacent cofferdam units in a variety of configurations to form watertight seals. The cofferdam units may also be sized and designed to attach to seawalls, flood walls, sidewalks, or other infrastructure. When in a deployed state, the height of the cofferdam may be adjusted to a desired height to provide flood mitigation. • Patent number: 11288953
Abstract: A method includes accessing a first dataset including aerial imagery data, accessing a second dataset including property boundary data, and identifying property boundaries associated with a geographic area. A plurality of artificial-intelligence (AI) models are applied to the datasets to identify and compute information of interest. Based on the first dataset and constrained by the property boundaries, a building detection model can be applied to identify a building footprint, and a tree detection model can be applied to identify one or more trees. An estimated distance can be determined between each of the trees and a nearest portion of the building footprint as separation data, which can be compared to a defensible space guideline to determine a defensible space adherence score. A wildfire risk map can be generated, including the defensible space adherence score associated with the geographic area.

  • Publication number: 20220074233
Abstract: A security system for a building includes a plurality of lock modules. Each lock module is associated with a particular door and has an unlocked position and a locked position. Each lock module includes a lock bar, an actuator, and a controller. The lock bar has a first position that allows the particular door to open and a second position that extends from a door frame, a wall, or a floor surface adjacent to the particular door to prevent the particular door from opening. The actuator moves the lock bar between the first and second positions. The controller is operably connected to the actuator and transmits a door status signal that uniquely identifies the particular door. A lock module command signal received by the plurality of lock modules directs the plurality of lock modules to reposition to either the unlocked position or the locked position.
