  • Publication number: 20250146053
Abstract: Electrode controlled hybridization is used to change local pH and selectively assemble oligonucleotide complexes on the surface of a microelectrode array. The oligonucleotide complexes have sticky ends that provide locations for subsequent oligonucleotide complexes to hybridize. The order in which specific oligonucleotide complexes are joined together encodes information. Controlled activation of individual electrodes in the microelectrode array creates negative voltages that reduces a buffer solution and raises the pH in proximity to the electrodes. At higher pH levels double-stranded oligonucleotides de-hybridize. Nicks between oligonucleotide complexes and oligonucleotides anchored to the microelectrode array are closed creating covalent attachments. De-hybridized single-stranded oligonucleotides are removed leaving only the oligonucleotides connected to microelectrode array.

  • Publication number: 20250150634
Abstract: Decoder for decoding a picture from a data stream, configured to check whether a plurality of coding parameters, which are contained in the data stream, relate to a predetermined portion of the picture and control a prediction residual transform mode and a quantization accuracy with respect to the predetermined portion, are indicative of a coding parameter setting corresponding to a lossless prediction residual coding. Responsive to the plurality of coding parameters being indicative of the coding parameter setting corresponding to the lossless prediction residual coding, the decoder is configured to set one or more predetermined coding options relating to one or more tools of the decoder for processing a prediction residual corrected predictive reconstruction with respect to the predetermined portion so that the one or more tools are disabled with respect to the predetermined portion.

  • Publication number: 20250115635
Abstract: This disclosure provides electrochemically-cleavable linkers with cleavage potentials that are less than the redox potential of the solvent in which the linkers are used. In some applications, the solvent may be water or an aqueous buffer solution. The linkers may be used to link a nucleotide to a bound group. The linkers include a cleavable group which may be one of a methoxybenzyl alcohol, an ester, a propargyl thioether, or a trichloroethyl ether. The linkers may be cleaved in solvent by generating an electrode potential that is less than the redox potential of the solvent. In some implementations, an electrode array may be used to generate localized electrode potentials which selectively cleave linkers bound to the activated electrode. Uses for the linkers include attachment of blocking groups to nucleotides in enzymatic oligonucleotide synthesis.

  • Patent number: 12265853
Abstract: Arranging computational sub-tasks in a hybrid-computing environment is provided. The method comprises receiving input of a number of nodes, wherein each node represents a computational sub-task, and wherein the nodes are grouped into different sets according to differing computing resources used by the nodes. A computational objective is also received as well as initial data inputs and desired final outputs. A directed graph network is generated comprising the nodes and directed edges connecting the nodes. An optimization problem is then solved to determine a best path through the directed graph network for deriving the desired final outputs from the initial data inputs according to the computational objective. The best path comprises a subset of nodes and directed edges within the directed graph network. The best path to achieve the computational objective and estimated computational resources required for the best path are output to a user.

  • Publication number: 20250092082
Abstract: Photon generating substrates for light-directed oligonucleotide synthesis are disclosed. Light is generated within a solid-state stack that supports growing oligonucleotides. The light may be generated by microLEDs, a pass-through liquid crystal panel, or an LCoS system. Light passes through a transmissive layer on which growing oligonucleotides are attached. Patterning of the light is controlled by selective activation of the microLEDs or by selective control of the transparency of a liquid crystal layer. Photolabile blocking groups are selectively removed by exposure to patterned light emitted from the photon generating substrate.

  • Patent number: 12252739
Abstract: Selectively controllable cleavable linkers include electrochemically-cleavable linkers, photolabile linkers, thermolabile linkers, chemically-labile linkers, and enzymatically-cleavable linkers. Selective cleavage of individual linkers may be controlled by changing local conditions. Local conditions may be changed by activating electrodes in proximity to the linkers, exposing the linkers to light, heating the linkers, or applying chemicals. Selective cleaving of enzymatically-cleavable linkers may be controlled by designing the sequences of different sets of the individual linkers to respond to different enzymes. Cleavable linkers may be used to attach polymers to a solid substrate. Selective cleavage of the linkers enables release of specific polymers from the solid substrate. Cleavable linkers may also be used to attach protecting groups to the ends of growing polymers. The protecting groups may be selectively removed by cleavage of the linkers to enable growth of specific polymers.

  • Patent number: 12206247
Abstract: A power output control and compensation (PCC) circuit operates within a distributed energy resource (DER) system. The PCC circuit is located in a battery power conversion system (PCS) and includes an input terminal to connect with a power source and an output terminal to connect with an electrical panel or load. A current sensing device is coupled between the input and the output terminals to measure a power level of the power source. A relay is coupled between the current sensing device and the output terminal and a processor selectively connects power from the power source to the electrical panel or the load based on the measured power level or based on other power measurements within the DER system. PCS-control of multiple power sources decreases cycle times for controlling distributed energy resources.

  • Patent number: 12198685
Abstract: Methods and systems are presented for translating informal utterances into formal texts. Informal utterances may include words in abbreviation forms or typographical errors. The informal utterances may be processed by mapping each word in an utterance into a well-defined token. The mapping from the words to the tokens may be based on a context associated with the utterance derived by analyzing the utterance in a character-by-character basis. The token that is mapped for each word can be one of a vocabulary token that corresponds to a formal word in a pre-defined word corpus, an unknown token that corresponds to an unknown word, or a masked token. Formal text may then be generated based on the mapped tokens. Through the processing of informal utterances using the techniques disclosed herein, the informal utterances are both normalized and sanitized.

  • Patent number: 12197834
Abstract: A method of minimizing a cost function of a quantum computation is provided. The method comprises receiving input of an initial state of a quantum problem instance comprising a Hamiltonian with an associated cost function. The Hamiltonian is converted into a number of Pauli strings, which are used to form an operator pool. The Pauli strings in the operator pool are ranked according to how much they lower a value of the cost function with respect to the initial state. Pauli strings are iteratively added from the operator pool to a parameterized quantum circuit, in a manner to minimize circuit depth, until a variational quantum eigensolver (VQE) algorithm converges to an approximate ground state wave function generated by the parameterized quantum circuit.
