  • Publication number: 20220265865
Abstract: The disclosure features compositions and methods for the treatment of sensorineural hearing loss and auditory neuropathy, particularly forms of the disease that are associated with a mutation in otoferlin (OTOF), by way of OTOF gene therapy. The disclosure provides a variety of compositions that include a first nucleic acid vector that contains a polynucleotide encoding an N-terminal portion of an OTOF isoform 5 protein and a second nucleic acid vector that contains a polynucleotide encoding a C-terminal portion of an OTOF isoform 5 protein. These vectors can be used to increase the expression of OTOF in a subject, such as a human subject suffering from sensorineural hearing loss.

  • Publication number: 20210388045
Abstract: The disclosure provides polynucleotides containing regions of the Myosin 15 (Myo15) promoter, as well as vectors containing the same, that can be used to promote expression of a transgene specifically in hair cells. The polynucleotides described herein may be operably linked to a transgene, such as a transgene encoding a therapeutic protein, so as to promote hair cell-specific expression of the transgene. The polynucleotides described herein may be operably linked to a therapeutic transgene and used for the treatment of subjects having or at risk of developing hearing loss or vestibular dysfunction.

  • Publication number: 20210307286
Abstract: A method and apparatus (3) for operating a rotary milking platform (1) to maximise the number of animals milked per unit time. The milking platform (1) is rotated about a central vertical axis (4) by a variable speed motor (6), and comprises a plurality of animal accommodating locations (5) for the animals being milked. An entry position (7) and an exit position (9) accommodate animals to and from the platform (1). A position sensor (10) monitors the angular position of the platform (1). An RFID sensor (12) reads the identity of animals entering the platform (1). Historical data relating to milking time per milking session and the milk yield per animal per session is stored in an electronic memory (17). A microprocessor (15) reads signals from flow meters (14) which monitor the milk flow from milking clusters of each animal accommodating location (5).

  • Patent number: 10321975
Abstract: A method for continuously evaluating the effectiveness of debridement of a root canal of a tooth, the tooth having an open access cavity and an apex end, includes delivering a fluid to the open access cavity of the tooth, evacuating the fluid near the apex end of the tooth such that the fluid flushes most of the root canal before being evacuated, and continuously evaluating the evacuated fluid for at least one of a presence of debris, a concentration level of the debris, or a type of the debris. An apparatus for use in debriding a root canal of a tooth includes a microcannula or a macrocannula configured to evacuate a fluid in the root canal, and a sensing mechanism fluidically coupled to the microcannula or the macrocannula, the sensing mechanism configured to continuously sense debris in the evacuated fluid in real time.

  • Publication number: 20150290778
Abstract: A multifunctional tool having an interchangeable head is provided. The tool includes a pair of arms each having a jaw displacement end. A female pivot member having a central aperture is interdisposed between the jaw displacement ends and pivotally connects to each arm. At least two nodes are disposed on the front face and rear face of the jaw displacement end. An interchangeable head having a plurality of apertures and a male pivot member mates with the nodes of the female pivot member disposed on the jaw displacement end. A locking pin extends through the central apertures of the pivot members to attach the interchangeable head to the jaw displacement ends. When the interchangeable head is attached to the nodes on the front face, the tool functions as a clamp. When the interchangeable tool is attached to the nodes on the rear face, the tool functions as a separating apparatus.
