  • Publication number: 20200271564
Abstract: A method of sorting cells including magnetically coupling a nozzle assembly to a flow cytometer in a predetermined orientation to form a fluid stream having a sheath fluid and a sample fluid. Particles within the sample fluid are oriented and then interrogated at an inspection zone to produces signals representative of emitted or reflected electromagnetic radiation. The electromagnetic radiation is analyzed to classify the cells and the cells are sorted according to their classification.

  • Publication number: 20200200762
Abstract: Cell sorting methods that improve sorting efficiency and productivity by elevating sorting pressures and incorporate certain steps to help the cells better survive such elevated pressures. In the case of sperm, sorting the steps of standardizing sperm samples, staining sperm samples in a single step, calibrating a flow cytometer to place sperm in the leading edge of droplets, and changing a catch fluid distance may be incorporated individually, or in combination to help sperm better survive the sex sorting process.

  • Publication number: 20200158622
Abstract: In flow cytometry, particles (2) can be distinguished between populations (8) by combining n-dimensional parameter data, which may be derived from signal data from a particle, to mathematically achieve numerical results representative of an alteration (48). An alteration may include a rotational alteration, a scaled alteration, or perhaps even a translational alteration. • Patent number: 9581587
Abstract: A method of sex sorting sperm is disclosed. The sperm may be stained with a DNA selective fluorescent dye, which fluoresces when excited, a dead quenching dye, which selectively quenches fluorescence emitted by the DNA selective fluorescent dye within the membrane of compromised sperm, and a split enhancing dye. The stained sperm may then sorted into one or more gender enriched subpopulations of viable sperm.

Abstract: Embodiments of the claimed invention relate to a method of producing sex sorted sperm having an improved dose response which includes the steps of extending a sperm sample with a buffering holding media and adjusting the concentration of the extended sperm sample to a target concentration range. The extended sperm may then be stained with a DNA selective dye and sex sorted with a flow cytometer into a catch fluid. During the extending and sorting of the sperm sample, the pH may be maintained at a target pH. Additionally, at least one of the buffering holding media, the DNA selective dye and the catch fluid may be supplemented with at least one antioxidant.
