    Abstract: Methods and compositions for detection of microbial contaminants in peritoneal dialysis solutions are provided. The methods and compositions employ modified bioburden testing and the detection of peptidoglycan. A novel cause of aseptic peritonitis is provided—aseptic peritonitis associated with gram positive microbial contamination of a dialysis solution. Peptidoglycan is a major component of a gram positive bacterial cell wall and thus can serve as a marker for gram positive bacteria. In this regard, testing for peptidoglycans can be utilized to effectively prevent peritonitis in patients that use the peritoneal dialysis solutions, such as peritoneal dialysis solutions that contain a glucose polymer including an icodextrin and the like.

    Abstract: Peritoneal dialysis solutions including a glucose polymer and methods of using the dialysis solutions are disclosed herein. In a general embodiment, the peritoneal dialysis solution includes a glucose polymer in an amount to provide an increased ultrafiltration fluid volume for a given amount of carbohydrate absorbed compared to conventional dialysis solutions containing icodextrin as the active pharmaceutical ingredient. The increased ultrafiltration fluid volume for a given amount of carbohydrate absorbed is obtained by providing specific combinations of weight-average molecular weight, polydispersity index and concentration of the glucose polymer in the dialysis solution.

    Abstract: In a peritoneal dialysis embodiment of the present invention, spent dialysate from the patient's peritoneal cavity passes, along a patient loop, through a dialyzer having a membrane that separates waste components from the spent dialysate, wherein the patient loop returns fresh dialysate to the patient's peritoneal cavity. The waste components are carried away in a second regeneration loop to a regeneration unit or sorbent cartridge, which absorbs the waste components. The regeneration unit removes undesirable components in the dialysate that were removed from the patient loop by the dialyzer, for example, excess water (ultrafiltrate or UF), toxins and metabolic wastes. Desirable components can be added to the dialysate by the system, such as glucose and electrolytes. The additives assist in maintaining the proper osmotic gradients in the patient to perform dialysis and provide the necessary compounds to the patient.
