  • Patent number: 6689584
Abstract: BLAST search was done on the EST database by using various nucleotide sequences encoding known bromodomain motifs to discover several ESTs likely encoding bromodomain genes. Next, testicular cDNAs were PCR cloned by using primers designed based on the sequence of EST (W17142), which is one of the ESTs discovered above. By using the thus obtained PCR product as a probe, the testicular library was screened. The obtained cDNA clone was used as a probe to re-screen the testicular cDNA library, thereby successfully isolating a full-length cDNA corresponding to EST (W17142). The protein encoded by the thus isolated cDNA had, in addition to the bromodomain, several regions and domains conserved in transcription regulatory factors. Moreover, the protein interacted with proteins associated with the chromatin-mediated transcriptional regulatory mechanism and a transcription co-activator.

  • Publication number: 20030083470
Abstract: A database search using the sequence of transcriptional regulator “RING3” having a bromodomain identified an EST sequence that is highly homologous with RING3. Using primers prepared based on the EST sequence, a gene encoding a novel transcriptional regulator having a bromodomain has been successfully isolated from a human testis cDNA library by polymerase chain reactions. The results of analysis of the isolated gene revealed that this gene is expressed strongly in testis cells with a potent proliferative ability. The use of the above transcriptional regulator and its gene enables screening candidate compounds for factors interacting with the transcriptional regulator or drugs controlling the activity of the regulator.

  • Patent number: 4624395
Abstract: A hot beverage dispensing machine is disclosed for dispensing a beverage from a concentrate. The dispensing machine includes a beverage concentrate container disposed in an inverted orientation within a refrigerated compartment. A first valve is associated with the container, the first valve permitting the flow of the concentrate from the container when the container is located within the refrigerated compartment in an inverted position. A receptacle having a receptacle output removably receives the inverted container. A first electrically controlled valve regulates the flow of the concentrate via the receptacle output to a mixer wherein the concentrate is mixed with hot water from a hot water tank. A second electrically controlled valve regulates the flow of hot water from the water tank to the mixer. A dispenser spout is disposed in fluid communication with the mixer with the spout being disposed remote from the hot water inlet.

  • Patent number: 4416604
Abstract: A universal injection molding system has plural pairs of mold inserts that are engageable for mold cavities mounted on mold bases, respectively, on the relatively movable top clamp plate and base support plate of an injection molding machine. Rails on those bases hold the mold inserts in place in paired alignment. With the plates and bases closed to bring together respective mold insert pairs, the mold inserts are securely clamped in relatively fixed positions on the plates and bases. A multiple stage ejecting/stripping apparatus also is provided to strip parts from mold cavities, and a technique is provided to eject the sprue before the parts. Methods of facilitating set-up of an injection molding system and of injection molding also are disclosed.
