The one-pot biosensor may detect a target by once solution injection and has a size enough to be portable. Accordingly, since the one-pot biosensor can detect the target by only once solution injection without a washing step, because of a sensor platform capable of being easily used by an individual other than a diagnostic expert, it is predicted to be positioned as a means capable of confirming the health condition of the individual without seeing the doctor, such as a pregnancy diagnostic kit which has been currently commercialized.

    Abstract: The present invention relates to an aptamer screening method, and the aptamer screened by the screening method of the present invention binds to a site other than a site where the antibody binds to the target substance to be applicable in various fields such as sandwich-type biosensors and reduce a significant time without requiring a separate pairing selection process. In addition, such an aptamer has excellent stability and sensitivity compared to conventional preparations comprising an antibody, can be mass-produced at low cost in a short time by a chemical synthesis method, and is easily transformed in various ways to increase a binding force. In addition, the immunoassay method using the aptamer screened by the aptamer screening method of the present invention selectively amplifies only the aptamer binding to the target substance in a heterogeneous sandwich structure to detect a relative fluorescence signal, thereby detecting the target substance sensitively and quickly.

The aptasensor includes a light emitting unit for selectively emitting one of white light and a laser, a sensor unit including a plurality of aptamers, a plurality of gold nanorods, and a silver mirror, a detector for analyzing a wavelength of inputted light, and an optical fiber for connecting the light emitting unit with the sensor unit, and connecting the detector with the sensor unit, wherein the light emitted from the light emitting unit is totally reflected in the optical fiber and irradiated to the sensor unit, and light reflected from the silver mirror of the sensor unit is irradiated to the detector. Accordingly, the aptasensor easily measures concentration of a target material in a sample using the optical fiber.
