    Abstract: An online system trains a constraint prediction machine-learning model using an action-based loss function. The action-based loss function computes a weighted sum of (1) an accuracy of the constraint prediction model in predicting whether a user's interaction complies with a set of constraints and (2) an action score representing a number of actions taken by users to determine whether the user's action complies with the set of constraints. The online system may apply this trained constraint prediction model to future interaction data received from third-party systems to predict whether user interactions with those third-party systems comply with the set of constraints.

    Abstract: A closed, automated and scalable stirred tank bioreactor platform, capable of sustaining high fold expansion of hPSCs is provided. hPSCs are expanded in a controlled bioreactor using perfused xeno-free media. Cell harvest and concentration are performed in closed steps. The hPSCs can be cryopreserved to generate a bank of cells or further processed as needed. Cryopreserved cells can be thawed into a 2D tissue culture platform or a 3D bioreactor to initiate a new expansion phase or be differentiated to the clinically relevant cell type. The expanded hPSCs express hPSC-specific markers, have a normal karyotype and the ability to differentiate to the cells of the three germ layers. This end-to-end platform allows large expansion of high quality hPSCs that can support the required cell demand for various clinical indications.

    Abstract: Described herein are engineered anti-IL-2 antibodies with modified amino acid sequences. The engineered antibodies would confer modified receptor binding specificity to an IL-2-anti-IL2 antibody complex, inhibiting the binding of IL-2 to CD25. The engineered anti-IL-2 antibodies would facilitate expansion of subsets of effector immune cells and decrease undesirable effects caused by IL-2. Thus, the engineered anti-IL-2 antibodies would be useful in treating disease such as cancer and infection.

    Abstract: The disclosure relates to devices and methods for the treatment of edema which devices use an intravascular edema pump with an anticoagulant. The invention provides devices and methods for treating edema that use an indwelling catheter to place an impeller in a blood vessel of a patient, near an outlet of a lymphatic duct. Operating the impeller creates a local depression in blood pressure, which promotes drainage of lymph from the lymphatic system. The catheter is also used to release an anticoagulant such as heparin to wash and lubricate the impeller. Specifically, the anticoagulant inhibits clotting, hemolysis, or thrombosis from occurring and interfering with smooth operation of the impeller.

Devices and methods of the invention further use a flow-restrictor in the circulatory system, upstream of an intravascular pump, to balance pressure changes induced by the pump and to compensate for downstream flow. The device may be provided as an indwelling, intravascular catheter with a mechanical pump such as an impeller and a selectively deployable restrictor such as an inflatable balloon. Congestive heart failure or edema is treated by \ operating the pump in an innominate vein and using the restrictor for flow compensation, to restrict the upstream flow and thus amplify or maintain pressure reduction at the lymphatic outlet.
