    Abstract: Embodiment of the present invention discloses a kind of methionine lyase and its encoding gene and biosynthetic method. According to the present invention, the gene encoding methionine lyase as shown in SEQ ID No. 1 is separated from the genome of C. rosea. Embodiment of the present invention further provides an efficient biosynthetic method of methionine lyase, comprising: (1) cloning gene (shown in SEQ ID No.1) encoding methionine degradation enzyme into a yeast expression vector to construct recombinant yeast expression vector; (2) transforming the recombinant yeast expression vector into Saccharomyces cerevisiae to obtain expression strain; (3) inducing the expression strain to express the methionine lyase, collecting induced strains, purifying expressed recombinant methionine lyase. Purity of recombinant methionine lyase prepared according to the present invention is above 90%, and its efficiency of degradating methionine can reach 0.53±0.0030 ?M MTL·h?1·mg protein?1.

The present invention discloses a method for constructing an expression plasmid pETM6R1-RBS-glmM-GGGS-glmS that contains a glmM and a glmS gene under regulation of ribosome binding sites. The recombinant plasmid was transformed into E. coli K4 to obtain a recombinant E. coli strain ZQ33 that can produce fructosylated chondroitin up to 3.99 g·L?1 by fed-batch fermentation in a 5-L fermentor, which was increased by 108.90% compared to that of the wild type strain.

    Abstract: Methods for dequeue optimizations in queues are performed by systems and apparatuses. The methods optimize dequeue operations using aggregation of expired messages enqueued in a queue and conditional iteration over enqueued messages based on the aggregation to service dequeue commands. Queues utilize page hierarchies such as root pages, index pages, and data pages. The aggregation of expired messages for pages in the queue determines the latest expired time for messages for a given page, and these latest expired times are stored in their respective pages, including data pages, index pages, and root pages, for use in the conditional iteration. The conditional iteration bypasses pages for which a latest expired time for all messages is prior to the current time when servicing dequeue requests for the queue.

Additionally, the system can use this aggregate information internally for improved load balancing, better data placement, optimization, and enhanced debugging. The system can quickly look at aggregate information across a huge amount of data and drill down cheaply because the aggregate information is generated using existing processes. Aggregated statistics storage and collection may be built on top of an LSM tree used to store a persistent index for a cloud storage system. The statistics may also represent the result of an operation (e.g., max, min, sum, average) on selected parameter(s) or attribute(s) of stored data. Aggregate statistics values may be efficiently maintained during index merge and garbage collection processes or any other index management. As delta LSM trees are merged into a base LSM tree, the aggregates are updated in delta fashion.

The cathode is prepared by one-time microwave sintering. One-time forming of cathode sintering is realized, and sintering shrinkage and sintering time are reduced significantly. The method has excellent repeatability, and the cathode has a homogeneous structure and excellent emission performance at 950° C.
