    Abstract: The present invention provides fusion proteins for targeted delivery of plasminogen activators to platelet-VWF complexes, or alternatively to the site where these are located, in a fibrin-independent manner. The fusion protein of the invention are for use in methods for the prevention or treatment of diseases or conditions associated with such platelet-VWF complexes, which may cause microvascular thrombosis in diseases such as e.g. thrombotic thrombocytopenia purpura. Preferred targeting agents for incorporation into the fusion proteins are e.g. nanobodies against VWF or platelets. Preferred plasminogen activators for use in the fusion proteins comprise the protease domains of uPA or tPA. The invention further pertains to nucleic acid molecule encoding the fusion proteins of the invention, e.g. a gene therapy vector, and to pharmaceutical compositions comprising the fusion proteins of the invention or such gene therapy vectors.

    Abstract: A high strength J-bolt for a saddle-mount is provided for use in attaching the saddle-mount to an axle of a vehicle. The high strength J-bolt can include a round distal end, a tapered and/or round mid-section, and a hook end with a specified inside bend radius and cross-sectional shape configured to accommodate an increased range of axle sizes. A D-shaped cross-sectional shape can be formed for the hook end to provide for increased contact area with the axle, a larger cross-sectional area, and a higher section modulus along a critical bending axis as compared to a non-critical bending axis to maximize a clamping force that can be applied to the axle and militate against mechanical failure of the high strength J-bolt. A hot forging process can be used to form the shape of the high strength J-bolt.

According to some aspects, mechanisms for depositing and leveling source material are combined with a mechanism for heating the material. In some embodiments, one or more heating elements may be arranged to lead and/or follow a material deposition mechanism such that heat may be applied to the build region in concert with deposition of material. As a result of this technique, the heating and depositing steps may be performed closer together in time and/or heat may be applied more directly to the material than in conventional systems. As a result, greater control over material temperature may be achieved, thereby avoiding excess temperature exposure and subsequent undesirable changes to the material.
