  • Publication number: 20070130654
Abstract: Linoleic acid is converted into ?-linolenic acid by the enzyme ?6-desaturase. The present invention is directed to isolated nucleic acids comprising the ?6-desaturase gene. More particularly, the isolated nucleic acid comprises the promoter, coding region and termination regions of the ?6-desaturase gene. The present invention provides recombinant constructions comprising the ?6-desaturase coding region in functional combination with heterologous regulatory sequences. The nucleic acids and recombinant constructions of the instant invention are useful in the production of GLA in transgenic organisms.

  • Publication number: 20070059778
Abstract: Improved colony forming cell (CFC) assays are described. The improved assay comprises modifications to the standard CFC assay that enable analysis of temporal, real-time changes in antigen expression during colony development without need to fixate or destroy the culture. The improved assay is applicable to hematopoietic CFC assays as well as to CFC assays for other cells, such as neural cells and mammary cells. In one embodiment, the invention comprises adding a detection reagent, most likely a fluorescently labeled antibody, that is specific for antigens (most likely a cell-surface antigen) expressed on progenitors or on specific mature cell types to a culture at the start or during culture. The invention also comprises modifications to the culture medium and cell preparations used in standard CFC assays to selectively promote the development of one colony type while preventing or suppressing the development of other colony types.

  • Publication number: 20070020151
Abstract: A pipette tip holder is provided for holding a plurality of pipette tips in an automated pipette machine. The pipette tip holder includes a container that defines a cavity. There is at least one vertical divider in the cavity that divides the cavity into a plurality of segregated compartments. The pipette tip holder further includes a tip support member. The tip support member is connected to the container, The tip support member defines a plurality of apertures. The apertures are sized to receive and suspend said pipette tips in the cavity. Each aperture is generally aligned with one of said compartments. The at least one vertical divider is sized and positioned to inhibit contact between bottom portions of adjacent pipette tips.

  • Publication number: 20070016979
Abstract: The invention is directed to isolated promoters from stem-regulated, defense-inducible genes, such as JAS promoters. The promoters are useful in expression cassettes and expression vectors for the transformation of plants. Particularly, the invention provides transgenic plants of rice and sugarcane that have been modified such that expression of a heterologous coding sequence is directed by an JAS promoter and is limited to stem tissues or may be upregulated by the presence of a defense-inducing agent. The invention also discloses methods for producing the expression vectors and transgenic plants.
