Genes expressed after the three kinds of plasmids are transformed into protoplasts metabolizes a substrate L-DOPA, causing protoplast cells to produce betacyanin or betaxanthin that are visible to the naked eye. The two pigments are visible to the naked eye under an optical microscope and may be used to intuitively determine whether a protoplast transformation experiment is successful. The plasmids can stably indicate successfully transformed cells. Their self-luminous intensities are superior to intensities of red fluorescent proteins tdtomato and EGFP, and can perfectly replace the role of the fluorescent proteins in evaluating protoplast transformation efficiency.
