    Abstract: Provided are a helicase mutant, a preparation method therefor, and use thereof in high-throughput sequencing. The helicase mutant has at least one cysteine on a surface of a three-dimensional structure of an amino acid sequence as set forth in SEQ ID NO: 1, the at least one cysteine being substituted with alanine. The cysteine is C133, C164, C292, C323, and/or C347. Preferably, the amino acid sequence of the helicase mutant is as set forth in SEQ ID NO: 6. The helicase mutant exhibits excellent DNA unwinding activity, good protein uniformity, reduced cross-linking interactions between protein molecules, enhanced adaptability with perforin, improved sequencing stability, as well as integrity and continuity of electrical signals. Consequently, it achieves a better sequencing performance. It can be utilized for the control and characterization of nucleic acids and applied in single-molecule nanopore sequencing.

    Abstract: An online concierge system receives an order from a user including items to obtain from a retailer for delivery to a location. A picker selects the order and obtains items from the retailer. The user selects a time interval during which items from the order are delivered to the location. To prevent the user from selecting a time interval for fulfillment the online concierge system prevents the user from selecting a time interval when a picker may be unable to obtain the items from the retailer before a closing time of the retailer. The online concierge system evaluates time intervals by subtracting a travel time for the picker travelling from the retailer to the location from a predicted fulfillment time for the order. This prevents the time for delivering items after being obtained from affecting whether a time interval may be selected.

    Abstract: A comprehensive recycling method for a waste lithium iron phosphate battery relates to a waste lithium ion battery recycling technology, and particularly comprises: first selectively extracting lithium, and then using a lithium extraction residue to prepare iron phosphate, the using the lithium extraction residue to prepare the iron phosphate comprising: adding the lithium extraction residue to water to form a slurry, adding hydrochloric acid and stirring to react, so that iron is completely dissolved, performing solid-liquid separation, on the basis of iron and phosphorus contents of the obtained liquid, adding trisodium phosphate or ferric chloride, and then adding a sodium hydroxide solution to precipitate crude iron phosphate; and then performing reverse three-stage washing to remove impurities to obtain a battery iron phosphate product. The problem of environmental protection is solved and meanwhile, all of the valuable elements are recycled, and a relative cost is greatly reduced by about 25%.

A machine learning model, trained on historical data about available shoppers in discrete time intervals, is used to predict how many shoppers will be available to fulfill orders in each time interval. For each time interval, the system retrieves the relevant orders of both types and creates candidate groups including orders of both types. For each group, the system determines a fulfillment cost based on items in the orders. The candidate group with the lowest cost is selected, and the orders in the selected group are sent to devices of available shoppers in that interval, prompting the shoppers to view and fulfill the orders.
