Publication number: 20030207342Abstract: The present invention provides a method and a reagent for the simple and sensitive determination of cholesterol in remnant-like particles in a biological sample without the separation of components of the sample. The method comprises contacting the sample with (i) cholesterol esterase, (ii) cholesterol oxidase or cholesterol dehydrogenase, and (iii) phospholipase in the presence of oxygen or an oxidized coenzyme, and measuring the formed hydrogen peroxide or reduced coenzyme. The reagent comprises (i) cholesterol esterase, (ii) cholesterol oxidase or cholesterol dehydrogenase, and (iii) phospholipase.Type: ApplicationFiled: May 6, 2003Publication date: November 6, 2003Applicant: KYOWA MEDEX CO., LTD.Inventor: Kazuhito Miyauchi

Publication number: 20030207344Abstract: Ethers and esters of cyanohydrins that contain optically detectable moieties are highly effective for detecting, monitoring, and measuring the activity of enzymes that cause the cleavage of certain types of substrates. The cyanohydrins function as proaldehydes and proketones, spontaneously converting to aldehydes and ketones, respectively, which provide a large increase in optical detectability relative to both the starting esters and ethers and the cyanohydrins.Type: ApplicationFiled: January 7, 2003Publication date: November 6, 2003Applicant: THE REGENTS OF THE UNIVERSITY OF CALIFORNIAInventors: Bruce D. Hammock, Guomin Shan, Rong Zhang

Publication number: 20030207345Abstract: A method for detecting the presence of an oxygenated compound which is produced when a substrate is reacted with an oxygenase for the substrate. The method involves reacting a coupling enzyme with the oxygenated compound to form a polymeric oxygenated compound which is fluorescent or luminescent. Measurement of the fluorescence or luminescence of the polymeric oxygenated compound provides indirect detection of the oxygenated compound produced by reaction of the oxygenase with the substrate. The method is carried out in a whole cell environment wherein the cell is transformed to express both the oxygenase being screened and the coupling enzyme. The method can be used to measure the activity of monooxygenases and dioxygenases on aromatic substrates. The method is amenable to large scale screening of enzyme mutants to isolate those with maximum oxygenase activity.Type: ApplicationFiled: June 2, 2003Publication date: November 6, 2003Applicant: California Institute of TechnologyInventors: Frances H. Arnold, Hyun Joo

Publication number: 20030207346Abstract: The invention relates to a method of preparing heteromultimeric polypeptides such as bispecific antibodies, bispecific immunoadhesins and antibody-immunoadhesin chimeras. The invention also relates to the heteromultimers prepared using the method. Generally, the method provides a multispecific antibody having a common light chain associated with each heteromeric polypeptide having an antibody binding domain. Additionally the method futher involves introducing into the multispecific antibody a specific and complementary interaction at the interface of a first polypeptide and the interface of a second polypeptide, so as to promote heteromultimer formation and hinder homomultimer formation; and/or a free thiol-containing residue at the interface of a first polypeptide and a corresponding free thiol-containing residue in the interface of a second polypeptide, such that a non-naturally occurring disulfide bond is formed between the first and second polypeptide.Type: ApplicationFiled: August 12, 1999Publication date: November 6, 2003Inventors: WILLIAM R. ARATHOON, PAUL J. CARTER, ANNE M. MERCHANT, LEONARD G. PRESTA

Publication number: 20030207391Abstract: This invention relates to novel compounds that act to prevent dimerisation of vascular endothelial growth factor (VEGF) receptors. The novel compounds may comprise the amino acid sequence of the fourth Ig-like domain of a VEGF receptor, or a variant that retains the ability to bind to a VEGF receptor. These compounds are useful in the inhibition of the biological activity of VEGF receptors and may thus be used to treat diseases in which VEGF plays a role.Type: ApplicationFiled: June 7, 2002Publication date: November 6, 2003Inventor: Helen Pappa
