    Abstract: The present invention relates to a medium composition for enhancing the self-renewal ability of stem cells, which contains apelin, and a method of culturing stem cells using the medium composition. According to the present invention, stem cells can be effectively cultured to proliferate without changing the characteristics of the cells, and the telomerase activity of stem cells can be increased. Thus, the efficacy of cell therapy based on stem cells can be significantly improved.

More specifically, the invention relates to reverse transcriptases expressed and purified in bacteria and having the amino acid sequence of the reverse transcriptase of a human immunodeficiency virus type 1 (HIV-1) group O, modified at positions 358, 359 and 360; and variants of this enzyme that contain additional changes at positions 355 and 357 or at 478 or position 69 (in this case accompanied by an insertion of two amino acids). These polymerases have greater activity than the non-mutated enzyme at high temperatures (above 60° C.). In addition, they retain the capacity for DNA synthesis at temperatures greater than 70° C.     Abstract: Increases in antibiotic resistant strains of Staphylococcus aureus have elicited efforts to develop novel antimicrobials. One potential treatment includes lytic enzymes produced by staphylococcal bacteriophage during the lytic cycle. The phage Twort endolysin (PlyTW) harbors three domains, a CHAP endopeptidase, an amidase-2 domain, and a SH3b-5 cell wall binding domain. The CHAP domain alone is necessary and sufficient for lysis of live S. aureus; the amidase-2 domain alone is insufficient. Loss of the SH3b cell wall binding domain results in a 10 fold reduction of enzymatic activity in turbidity reduction and plate lysis assays compared to the full length protein. Deletion of the amidase-2 domain resulted in a protein (PlyTW ?172-373) with lytic activity that exceeded the activity of the full length construct in both assays. Addition of Ca2+ enhanced activity in turbidity reduction assays. Chelation by the addition of EDTA or zinc inhibited the activity of all PlyTW constructs.
