    Abstract: In one aspect, the invention provides methods of identifying genetic mutations that are associated with ataxic neurological disease. The methods comprise identifying a difference between a nucleic acid sequence of a protein kinase C gamma gene from a mammalian subject exhibiting ataxia and a nucleic acid sequence of a protein kinase C gamma gene from a subject which is not exhibiting ataxia, wherein the difference is a genetic mutation associated with ataxic neurological disease. In another aspect, isolated nucleic acid molecules encoding protein kinase C gamma missense mutations are provided. In another aspect, a method of screening a subject to determine if the subject has a genetic predisposition to develop an ataxic neurological disease is provided.     Abstract: The present methods are exemplified by a process in which maternal blood containing fetal DNA is diluted to a nominal value of approximately 0.5 genome equivalent of DNA per reaction sample. Digital analysis is then be used to detect aneuploidy, such as the trisomy that causes Down Syndrome. Since aneuploidies do not present a mutational change in sequence, and are merely a change in the number of chromosomes, it has not been possible to detect them in a fetus without resorting to invasive techniques such as amniocentesis or chorionic villi sampling. Digital amplification allows the detection of aneuploidy using massively parallel amplification and detection methods, examining, e.g., 10,000 genome equivalents.

    Abstract: Recent large-scale analyses have demonstrated that the genomic landscape of human cancer is complex and variable among individuals of the same tumor type. Such underlying genetic differences may in part be responsible for the varying therapeutic responses observed in cancer patients. To examine the effect of somatic genetic changes in colorectal cancer on sensitivity to a common targeted therapy, we performed complete exome sequence and copy number analyses of 129 tumors that were KRAS wild-type and analyzed their response to anti-EGFR antibody blockade in patient-derived tumorgraft models. In addition to previously identified genes, we detected mutations in ERBB2, EGFR, FGFR1, PDGFRA, and MAP2K1 as potential mechanisms of primary resistance to this therapy. Alterations in the ectodomain of EGFR were identified in patients with acquired resistance to EGFR blockade.

    Abstract: The invention relates to a thin sheet metal for a reinforcing or structural part of a car body, consisting of an aluminium alloy of the following composition, in weight per cent: Si: 10-14; Mg: 0.05-0.8; Cu: 0-0.2; Fe: 0-0.5; Mn: 0-0.5; optionally at least one element selected from Na, Ca, Sr, Ba, Yt and Li, the quantity of said element if selected being between 0.01 and 0.05 for Na, Ca, Sr, Ba and Yt and between 0.1 and 0.3 for Li; Sb: 0-0.05; Cr: 0-0.1; Ti: 0-0.2; other elements amounting to <0.05 each and a total of <0.15; and the remainder being aluminium. The invention also relates to the method for producing such a sheet metal and to the use of such a sheet metal for producing a reinforcing or strucural part for a car body. The sheet metals according to the invention advantageously have a modulus of elasticity of at least 77 GPa.
