Publication number: 20180346871Abstract: Methods and compositions herein provide non-naturally occurring ?-butyrolactones (GBLs) in racemic mixtures that increase efficiency and effectiveness of screening for production of antibiotics, and enhance yields and express silent pathways. Non-naturally occurring GBLs were synthesized and found to stimulate antibiotic production in several different streptomycete strains. Antibiotic production by Streptomyces coelicolor was induced by a racemic mixture of non-cognate stereoisomers of VB-D, seven of which are non-naturally occurring. Further, novel A-factor-type GBL analogs stimulated antibiotic production in S. coelicolor. Synthesis in response to the treatment with the non-cognates GBL was observed for known compounds including undecylprodigiosin, desferrioxamine and streptorubin B, as was synthesis of a compound of unknown structure.Type: ApplicationFiled: August 9, 2018Publication date: December 6, 2018Applicant: Prospective Research, Inc.Inventors: Dakota Benjamin Hamill, Jake J. Publication number: 20180346874Abstract: The invention provides for methods of obtaining cells from mammalian, preferably human, postpartum umbilical cord arterial tissue and use of these cells to treat ocular degenerative conditions. In particular, methods of the invention result in the isolation of a homogenous population of human postpartum umbilical cord arterial tissue-derived cells. In certain embodiments, the cells: express CD13, CD90, NG2, and HLA-ABC; do not express one or more of CD31, CD34, CD45, CD117, FSP1, and E-cadherin. In other embodiments, the cells secrete one or more of thrombospondin-1, BDNF and sVEGFR1.Type: ApplicationFiled: May 24, 2018Publication date: December 6, 2018Applicant: Janssen Biotech, Inc.Inventors: Ian R. Publication number: 20180346879Abstract: Methods and compositions for producing cells expressing CD56/Pax3, CD56Pax7, and/or Pax3/Pax7 are provided herein. In some instances, the method involves contacting a pluripotent stem cell in an in vitro culture with one compound, or with two or more compounds at the same time, wherein the contacting the pluripotent stem cell in the in vitro culture with one compound, or with two or more compounds at the same time, directly results in generation of cells expressing CD56/Pax3, CD56/Pax7, or Pax3/Pax7. Also provided are methods of using the generated cells. Typically one of the compounds is a wnt pathway activator, such as the GSK3 inhibitor CHIR99021.Type: ApplicationFiled: April 22, 2016Publication date: December 6, 2018Inventors: Uli SCHMIDT, Leslie CARON

Publication number: 20180346880Abstract: It is an object of the present invention to provide a method for producing frozen mesenchymal cells maintaining a viability and an adhesion rate similar to those of cells obtained through a step of culture, without undergoing a step of culturing cells before cryopreservation step. The method comprises dissociating cells by treating a tissue with proteolytic enzymes and cryopreserving the cells. The present invention provides a method for producing frozen mesenchymal cells maintaining a viability and an adhesion rate similar to those of culturing cells without a step of culturings cells before cryopreserving.Type: ApplicationFiled: May 2, 2016Publication date: December 6, 2018Inventors: Toshimitsu HIGA, Kenji OTANI, Koichi OTOMO
