Evanescent waves scattered from an object is used in a manner convenient to a user.

    Abstract: A prism system is disclosed for splitting a broadband incoming light beam. In one embodiment, the broadband beam enters the prism system and is shifted laterally through a parallelogram shaped prism. The beam then encounters a first dichroic coating treated surface. The light that is reflected off of the surface strikes the internal wall of the prism to undergo total internal reflection within the prism. The beam exits the prism parallel to the original incoming broadband beam. The beam that passes through the dichroic surface reaches a second dichroic coating treated surface where a specified waveband of light is reflected while the remaining wavelengths pass through. The beam reflected by the second dichroic surface undergoes a total internal reflection and exits parallel to the incoming broadband beam. The light beam which passes through the second dichroic surface exits the prism parallel to the original incoming broadband beam.

A dichroic beam combiner is used to combine a second laser beam with a first laser beam so that the two beams are collinear. A cylindrical lens with a vertically oriented axis in the focusable beam shaping optics can also be adjusted to adjust the location of the focus point of the two beams in the horizontal direction. Alignment is maintained with the opto-mechanical adjustments made on one laser beam relative to the other laser beam path. Additional beams can also be added to the optical path.

    Abstract: A method of controlling the illumination angle onto a target, including, illuminating onto the target with light from at least two light sources of pre-selected wavelengths; wherein each point on the target is illuminated by light from the light sources with a respective maximal illumination angle relative to a main illumination axis extending from substantially the center of the at least two light sources to the target, selecting a dichroic filter that transmits light from the at least two light sources as a function of the angle of incidence upon the filter, positioning the dichroic filter between the at least two light sources and the target to limit the transfer of light to light of the pre-selected wavelengths; and wherein the dichroic filter is selected to limit the maximal illumination angle illuminating each point on the target.

    Abstract: Methods and apparatus for standardizing quantitative measurements from a microscope system. The process includes a calibration procedure whereby an image of a calibration slide is obtained through the optics of the microscope system. The calibration slide produces a standard response, which can be used to determine a machine intrinsic factor for the particular system. The machine intrinsic factor can be stored for later reference. In use, images are acquired of a target sample and of the excitation light source. The excitation light source sample is obtained using a calibration instrument configured to sample intensity. The calibration instrument has an associated correction factor to compensate its performance to a universally standardized calibration instrument. The machine intrinsic factor, sampled intensity, and calibration instrument correction factor are usable to compensate a quantitative measurement of the target sample in order to normalize the results for comparison with other microscope systems.

    Abstract: Prism elements having TIR surfaces placed in close proximity to the active area of a SLM device to separate unwanted off-state and/or flat-state light from the projection ON-light bundle. The TIR critical angle of these prisms is selected to affect either the off-state light or additionally, any portion of flat-state light reflected from the SLM. These TIR surfaces limit the contamination of light along the projection path, which tends to degrade the system contrast. To further improve the optical performance of the system, these TIR prisms can be attached directly to the SLM package, completely eliminating the package window.
