In one procedure, the added drugs are pre-coupled to the microparticles while the antibodies are not, and the incubation is followed by further incubating the microparticles with labeled ligands that have affinity for the antibodies.

In one embodiment, this method occurs without a washing or physical cell separation step. The incubated sample is irradiated with a laser light source, and fluorescence of the labeled ligand and the nucleic acid dye are measured along with at least one additional parameter, e.g., light scatter, direct current, axial light loss, opacity, radio frequency, and fluorescence.

    Abstract: A new technique to stabilize transition metal phosphors in a wide variety of stimuli-sensitive polymers and gels is disclosed herein. Other than stabilization in stimuli sensitive/biocompatible matrix some of these transition metal based phosphors are also shown to act as phosphorescent crosslinkers that physically or chemically crosslink polymeric chains to form micro/nanoparticles. The microspheres/nanospheres of the present invention show decreased size and photoluminescence enhancement with particularly high sensitization at physiological pH and temperature. The so formed phosphorescent micro/nanospheres are useful for biological or environmental applications including biological labeling, imaging, and optical sensing. The techniques in the present invention enable usage of imaging agents and sensors at very low concentrations and also minimize or eliminate the usage of toxic chemical crosslinkers typically used to synthesize polymeric micro/nanoparticles.

A method of operating such a flow cytometer includes priming the interrogation flow cell with a sheath fluid and injecting different assay fluids into a flow of the sheath fluid through the plurality of fluidic lines. A fluidic line assembly is provided which includes a plurality of capillary tubes coupled to a base section configured for coupling to an interrogation flow cell assembly of a flow cytometer. The capillary tubes are dimensionally configured such that when the fluidic line assembly is arranged within the flow cytometer and fluid is dispensed from one or more of the capillary tubes at a given pressure differential with respect to an encompassing sheath fluid within the interrogation flow cell the fluid is substantially centrally aligned within the interrogation flow cell.
