Patent number: 5747256Abstract: An assay for detecting the presence or amount of a target polynucleotide sequence of interest in a test sample is disclosed. The assay comprises the steps of forming a reaction mixture by combining in an assay medium (i) a first reagent and (ii) an aliquot of the test sample suspected of containing the target polynucleotide sequence. The reaction mixture is subjected to denaturing conditions, and exposed to hybridization conditions allowing the first reagent to hybridize with any target polynucleotide present. A second reagent is then added, the reaction mixture undergoes hybridization conditions and the change in turbidity of the reaction mixture is detected, correlating with the presence of the target polynucleotide in the sample.Type: GrantFiled: December 19, 1995Date of Patent: May 5, 1998Assignee: Beckman Instruments, Inc.Inventors: Cheng F. Patent number: 5734468Abstract: A method and device for detecting the presence of hemolysis, icteris and lipemia in a serum sample are provided herein. The method includes the steps of positioning a probe inlet of a probe in a sample container, aspiring at least a portion of the serum sample into a probe lumen of the probe from the sample container, and monitoring the serum sample in a probe lumen to determine whether hemolysis, icteris and lipemia are present in the serum sample. The serum sample is monitored with a detector which performs spectrophotometric analysis of the serum sample in the probe lumen through a substantially transparent section of the probe. From the spectrophotometric analysis, a hemolytic index, an icteric index and a lipemic index of the serum sample can be established. Based upon these serum indices, the serum sample can be transferred to a clinical analyzer for additional tests or can be disposed of because the sample is compromised.Type: GrantFiled: November 12, 1996Date of Patent: March 31, 1998Assignee: Beckman Instruments, Inc.Inventor: Jack D. Patent number: 5667755Abstract: A hybrid fiber-composite centrifuge container having a cylindrical metal sleeve, with a smooth inner surface permanently coupled to the fiber reinforced base. The reinforced base has an open end, a closed end and a cylindrical wall extending between the closed and open ends, with the metal sleeve coupled to the cylindrical wall proximate to the open end forming a sleeve-base interface. The base is formed from a plurality of layers of fiber material wound helically and circumferentially about the axis of the base. Patent number: 5666048Abstract: A technique and circuit for measuring direct current flowing through a conductor at a high voltage employs a capacitor connected in series with the conductor, and a neon lamp connected in parallel with the capacitor. The series connected capacitor and conductor are connected to a high voltage source such that the current flowing through the series connected capacitor and conductor charges up the capacitor. Since the capacitor and the neon lamp are connected in parallel, the voltage across the electrodes of the neon lamp follows the voltage across the capacitor. When the voltage across the electrodes of the neon lamp reaches the lamp's ignition voltage, the neon lamp fires, and discharges the capacitor until the voltage across the electrodes of the neon lamp falls to the lamp's extinction voltage. The light signal generated by the neon lamp is picked up by a fiber optic cable and transmitted to a remote receiver for processing.Type: GrantFiled: June 26, 1995Date of Patent: September 9, 1997Assignee: Beckman Instruments, Inc.Inventors: Donald P. Patent number: 5662787Abstract: Methods for profiling oligosaccharides released from glycoproteins and methods for the electrophoretic separation of high mannose, complex and hybrid oligosaccharides are disclosed. The disclosed methods include first providing an analytical sample which includes at least one released oligosaccharide and introducing the sample into an electrophoretic channel containing an electrophoretic separation medium having a pH less than 8 and greater than 2.5.Then applying a sufficiently high electric field causes the released oligosaccharides to electrophoretically migrate and separate within the electrophoresis channel in accordance with their molecular size or charge to mass ratio. Detecting the separated oligosaccharides provides glycoprotein conjugate profiling information. Preferred embodiments include using an electrophoresis separation medium which includes a lithium acetate buffer having pH of about 4.75 and a gel of about 0.4 wt % polyethylene oxide.Type: GrantFiled: January 19, 1996Date of Patent: September 2, 1997Assignee: Beckman Instruments, Inc.Inventors: Andras Guttman, Nelson H. Patent number: 5661019Abstract: A trifunctional conjugate is providing having three chemical moieties attached through a spacer moiety. At least two of the chemical moieties are relatively small molecules, usually less than about 7,000 Daltons in size. The spacer moiety is selected to impart certain steric properties to the conjugate. In one embodiment, the binding of a macromolecular specific binding partner to one of the chemical moieties sterically inhibits the binding of a different macromolecule to another chemical moieties. In another embodiment, the binding of a first chemical moiety to a macromolecule restricts the subsequent binding of a second tridentate member to a proximate location on the same macromolecule. The three chemical moieties are preferably a nitrophenylazido residue, a phenyl boronic acid residue, and a solid support or a label such as biotin. The spacer is preferably cysteine, lysine, glutamic acid, pyroglutamic acid, S-acetylmercaptosuccinic anhydride or .omega.-carbobenzoxylysine.Type: GrantFiled: March 22, 1995Date of Patent: August 26, 1997Assignee: Beckman Instruments, Inc.Inventors: Chan S. Patent number: 5648213Abstract: Double stranded nucleic acid duplexes serve as universal harvestable and cleavable link systems in a variety of different types of immunoassays (e.g., sandwich, competitive, etc.). Depending upon the type of assay, at least one specific component involved in the assay system is attached to a first member of a pair of sequences forming a double stranded nucleic acid (i.e., two oligonucleotides comprising substantially complementary sequences). The assay is carried out in the presence of a support to which is attached an oligonucleotide which is the other member of the pair of sequences forming a double-stranded nucleic acid duplex under hybridization conditions. Upon the hybridization of the two complementary oligonucleotides to form a duplex, the component of the assay system to which the first member of the pair of oligonucleotides is attached may thereby be effectively removed from the solution phase and harvested onto the support.Type: GrantFiled: August 30, 1994Date of Patent: July 15, 1997Assignee: Beckman Instruments, Inc.Inventors: M. Patent number: 5635050Abstract: A gel pump operated by a stepper motor may be used to deliver fresh gel to one or more capillaries through one or more manifolds and valves. By controlling the valves, fresh gel delivered by the pump will replace the old gel in the capillaries for an automated gel replacement system. In a different setting of the valves, the manifolds may be purged of the old gel prior to delivery of fresh gel to the capillaries. The gel delivery system may also be combined with an electrophoresis system so that sealing connection capable of withstanding high pressure adequate for gel injection need not be frequently broken when the gel is to be replaced. Manifold/reflector assembly is advantageously used in the system that facilitates electrical circuit for electrophoresis and for gel replacement.Type: GrantFiled: August 23, 1995Date of Patent: June 3, 1997Assignee: Beckman Instruments, Inc.Inventors: Stephen L.
