    Abstract: A filtration device including a contoured support having 1) an interior surface, 2) an outside peripheral surface spaced farther from a center of the contoured support than the interior surface, and 3) a rim enclosed and sealed to a perimeter of the contoured support. The filtration device includes macroscopic openings in the contoured support extending from the interior surface to the outside peripheral surface, and a plurality of nanofibers having diameters less than 1 micron. The nanofibers are disposed on the outside peripheral surface of the contoured support, and cover the macroscopic openings to form a filtration medium on the contoured support.

    Abstract: HIV-1's ability to enter a transcriptionally dormant state and establish a reservoir of latently infected cells is considered the major barrier to eradicating the virus from infected patients. Stochastic noise (i.e. fluctuations) in an HIV-1 transcriptional positive-feedback loop is one mechanism that enables HIV-1 to establish latency. Here, Applicants demonstrate that small-molecule modulation of noise in HIV-1 gene expression radically perturbs HIV-1 latency.

    Abstract: Population-Hastened Assembly Genetic Engineering is a method for continuous genome recoding using a mixed population of cells. Nucleic acid donors are distributed amongst a population of cells that continuously transfer nucleic acids to achieve asynchronous recoding of genetic information within a subpopulation of the cells. Recombination is achieved with biochemical systems compatible with virtually any organism. An engineered directed endonuclease comprises a nucleic acid recognition domain, a nucleic acid endonuclease domain, and a linker fusing or causing interaction between the nucleic acid recognition domain and the nucleic acid endonuclease domain. The method includes causing at least one engineered directed endonuclease to create a nick in a nucleic acid strand, the nick being offset from the recognition sequence of the nucleic acid recognition domain; causing homologous recombination of the strand with a donor nucleotide to create a modified genome; and replicating the modified genome.
