  • Patent number: 10472650
Abstract: The present disclosure provides targeting peptides and vectors containing a sequence that encodes targetting peptides that deliver agents, to the eye. The present inventors have discovered peptides that function to target agents, such as viral vectors, to ocular cells. The present disclosure describes a method to utilize these novel peptides to direct, for example, viral capsids to the cell type of interest. In this instance, ocular cells (such as retinal cells) are targeted by the identified peptides. Vectors harboring capsid proteins modified to include such peptides can be used to provide therapeutic agents to the eye.

  • Patent number: 10471057
Abstract: Potent modulators of RNA function can be assembled in cellulo by using the cell as a reaction vessel and a disease-causing RNA as a catalyst. When designing small molecule effectors of function, a balance between permeability and potency must be struck. Low molecular weight compounds are more permeable while higher molecular weight compounds are more potent. The advantages of both types of compounds could be synergized if low molecular weight molecules could be transformed into potent, multivalent ligands via a reaction catalyzed by binding to a target in cells expressing a genetic defect. We demonstrate that this approach is indeed viable in cellulo. Small molecule modules with precisely positioned alkyne and azide moieties bind adjacent internal loops in r(CCUG)exp, the causative agent of myotonic dystrophy type 2 (DM2), and are transformed into oligomeric, potent inhibitors of DM2 RNA dysfunction via a 1,3 Huisgen dipolar cycloaddition reaction, a variant of click chemistry.

  • Patent number: 10472625
Abstract: The present invention relates to a Drosophila in vitro system which was used to demonstrate that dsRNA is processed to RNA segments 21-23 nucleotides (nt) in length. Furthermore, when these 21-23 nt fragments are purified and added back to Drosophila extracts, they mediate RNA interference in the absence of long dsRNA. Thus, these 21-23 nt fragments are the sequence-specific mediators of RNA degradation. A molecular signal, which may be their specific length, must be present in these 21-23 nt fragments to recruit cellular factors involved in RNAi. This present invention encompasses these 21-23 nt fragments and their use for specifically inactivating gene function. The use of these fragments (or chemically synthesized oligonucleotides of the same or similar nature) enables the targeting of specific mRNAs for degradation in mammalian cells, where the use of long dsRNAs to elicit RNAi is usually not practical, presumably because of the deleterious effects of the interferon response.

  • Patent number: 10470379
Abstract: Systems, methods, and apparatus for large scale, high throughput phenotyping of items including plants to derive environment/genetics correlations based on controlled variation of environment. Spatial and temporal resolution is improved by an integrated system that can use space efficiently yet concurrently process a large number of replicates but in varying environments. This is accomplished by using an array of miniature greenhouses, each in communication with one or more generators or regulators of an environmental factor that can be independently supplied and controlled to each miniature greenhouse. The controller also controls acquisition of data from each greenhouse as well as processing of that data into phenomic/genomic correlations.

  • Patent number: 10471261
Abstract: A connector block that permits simultaneous and continuous interconnection of the three leads of the epicardial pacing wires, the pacemaker, and the ECG monitor on clear separately labeled connectors is provided. Circuitry is provided that allows the display of epicardial signals on the telemetry unit, while still preserving the ability to pace the heart from the pacemaker. When pacing the connector block prevents excessive loading of the pacer signals by the ECG monitor and/or damage to the monitor by the high-voltage pacer signals. The connector block may be used universally on all monitors without the need for sophisticated understanding of the electrical characteristics of the ECG monitor or concern for damage or improper signal loading.
