    Abstract: To identify novel agents to treat carbapenem-resistant Acinetobacter baumannii. the Inventors used a nutrient-depleted medium with serum to mimic the in vivo environment. In RPMI with serum, the screen identified rifabutin (RBT) as being 133-fold more potent than rifampin (RIF) against A. baumannii, with MICs of 0.031 ?g/ml and 4 ?g/ml respectively. No difference in RBT vs. RIF activity was observed when MHII was used as the culture media. RBT possesses markedly superior in efficacy to RIF in murine models of lethal iv and pneumonia models of infection with a hyper-virulent, clinical lung and blood isolate of extreme-drug resistant (XDR) A. baumannii. In both models, RBT significantly improved survival and lowered bacterial burden compared to RIF. RBT is a promising, novel therapeutic option for A. baumannii infections.

    Abstract: Described herein are nanoparticle bound self-antigens as an immune and vaccine formulations to elicit self-regulations and reduce atherosclerosis. The use of apolipoprotein B100 (ApoB-100) peptide P210 was investigated in self-assembling peptide amphiphile micelles (P210-PAM) as a vaccine formulation to reduce atherosclerosis in ApoE?/? mice. Demonstrated herein, P210 provided T cell activation and memory response in peripheral blood mononuclear cells of human subjects with atherosclerotic cardiovascular disease, and dendritic cell uptake of P210-PAM and its co-staining with major histocompatibility complex class I (MHC-I) molecules supported its use as an immunogenic composition. In ApoE?/? mice, immunization with P210-PAM dampened P210-specific CD4+ T cell proliferative response and CD8+ T cell cytolytic response, modulated macrophage phenotype, and significantly reduced aortic atherosclerosis. P210-PAM immunization also reduced atherosclerosis in chimeric mice with human MHC-I allele.

    Abstract: Novel channel estimation techniques that can be performed solely in the analog domain are provided. The techniques use continuous analog channel estimation (CACE), periodic analog channel estimation (PACE), and multiantenna frequency shift reference (MAFSR). These techniques provide sufficient channel knowledge to enable analog beamforming at the receiver while significantly reducing the estimation overhead. These schemes involve transmission of a reference tone of a known frequency, either continuously along with the data (CACE, MAFSR) or in a separate phase from data (PACE). Analysis of the methods show that sufficient receiver beamforming gain can be achieved in sparse channels by building the beamformer using just the amplitude and phase estimates of the reference tone from each receiver antenna.

    Abstract: Current kidney organoids model development and diseases of the nephron but not the con-tiguous epithelial network of the kidney's collecting duct (CD) system. Here, we report the generation of an expandable, 3D branching ureteric bud (UB) organoid culture model that can be derived from primary UB progenitors from mouse and human fetal kidneys, or gen-erated de novo from human pluripotent stem cells. In chemically-defined culture conditions, UB organoids generate CD organoids, with differentiated principal and intercalated cells adopting spatial assemblies reflective of adult kidney's collecting system. Aggregating 3D-cultured nephron progenitor cells with UB organoids results in a reiterative process of branching morphogenesis and nephron induction, similar to kidney development. Applying a gene editing strategy to remove RET activity, we demonstrate genetically modified UB organoids can model congenital anomalies of kidney and urinary tract (CAKUT).

    Abstract: A lacrimal tear flow measurement device, and methods of manufacture and use, are described that includes a polymer microcapillary tube or similar structure having at least one end coated on the outside with soft silicone rubber and one end treated on the inside to be hydrophobic. The hydrophobic end keeps liquid from escaping or entering that end while allowing air to pass. The rest of the tube's insides may be hydrophilic or a neutral hydrophobe. As a Schirmer's test strip replacement, the entrance end of the device can be touched to the lacrimal lake of a patient's eye to collect suck up, or merely collect, tear fluid within the collection tube for weighing, volume measurement, or other analysis. Long-term collection devices for wear between doctors' visits can have a bypass channel allowing liquid to flow back onto the eye.
