Abstract: Nucleic acids encoding mutant elongation factor proteins (EF-Sep), phosphoseryl-tRNA synthetase (SepRS), and phosphoseryl-tRNA (tRNASep) and methods of use in site specific incorporation of phosphoserine into a protein or polypeptide are described. Mutant EF-Sep proteins are disclosed that bind Sep-tRNASep and protect Sep-tRNASep from deacylation. In a preferred embodiment the nucleic acids are on vectors and are expressed in cells such as bacterial cells, archeaebacterial cells, and eukaryotic cells. Proteins or polypeptides containing phosphoserine produced by the methods described herein can be used for a variety of applications such as research, antibody production, protein array manufacture and development of cell-based screens for new drug discovery.

Abstract: Described is a novel method for feature extraction for automatic gait recognition. This method uses Multi-kernel Fuzzy-based Local Gabor Binary Pattern. From a captured gait video sequence, the gait period is determined then a gait energy image is constructed to represent the spatial-temporal variations during one motion cycle of the gait sequence. Then, each gait sequence is represented with a feature vector. The computation of this vector is conducted by first applying the 2D Gabor filter bank then encoding the variations in the Gabor magnitude using a multi-kernel fuzzy local binary pattern operator. Finally, gait classification is performed using a support vector machine.

Abstract: The disclosed embodiments relate to a system that uses a video game to improve spatial and/or temporal information-processing capabilities of a user. During operation, the system enables the user to play the video game. During execution of the video game, the system first measures spatial and/or temporal information-processing capabilities of the user during the course of playing the video game. Next, the system uses the measured spatial and/or temporal information-processing capabilities to control a spatial placement and/or a temporal presentation rate of target items that the user is required to respond to during subsequent game play to stimulate enhancement of the user's spatial and/or temporal information-processing capabilities.

Abstract: The present invention discloses a device and methods for the analysis of secreted molecules from a single cell. As described herein, the invention incorporates individual microwells with a bottom surface capable of capturing a cell and allows the release of secreted molecules to be printed onto a capture surface. The device provides accurate identification of the cell source of the printed molecules by mapping the printed molecules to the cell source. The invention further employs spectrometry, immunoassay or label free Surface Plasmon Resonance imaging for detection of the secreted molecules in combination with a microwell array, where single cells are seeded in individual microwells and the secreted molecules are captured by the capture surface in an array print while the cell remains in the microwell for additional interrogation. The present invention has applications in medical research and diagnosis where individual target cells in a fluid sample are interrogated for the secreted products.
