    Abstract: A high temperature, high strength Ni—Co—Cr alloy possessing essentially fissure-free weldability for long-life service at 538° C. to 816° C. contains in % by weight about: 23.5 to 25.5% Cr, 15-22% Co, 1.1 to 2.0% Al, 1.0 to 1.8 % Ti, 0.95 to 2.2% Nb, less than 1.0% Mo, less than 1.0% Mn, less than 0.3% Si, less than 3% Fe, less than 0.3% Ta, less than 0.3% W, 0.005 to 0.08% C, 0.01 to 0.3% Zr, 0.0008 to 0.006% B, up to 0.05% rare earth metals, 0.005% to 0.025% Mg plus optional Ca and the balance Ni including trace additions and impurities. The strength and stability is assured at 760° C. when the Al/Ti ratio is constrained to between 0.95 and 1.25. Further, the sum of Al+Ti is constrained to between 2.25 and 3.0. The upper limits for Nb and Si are defined by the relationship: (% Nb+0.95)+3.32(% Si)<3.16.

A content provider may initially mark the content for advertisement insertion, and create a metadata file indicating constraints, targets, expirations, or the like. A downstream user employs a plug-in component and provides an initial user profile, in part, to access the content with advertisements. As the content is played, and an advertising marker is encountered, fast forwarding or other skipping features are disabled, and an advertisement stream is spliced into the content stream. The advertisement stream is determined based on the user profile and/or other metadata. Moreover, the advertisement stream may be obtained over a network such that advertisements may be refreshed even years after the content has been acquired by the user. In one embodiment, the advertisement consumption may be tracked and reported.

    Abstract: Molecular constructs and dual recognition constructs having a sequence encoding a TCR affinity weakening motif, and DNA and RNA sequences corresponding thereto, are presented. Modified T-cells and other cells transformed with the molecular contracts express a modified TCR that imparts a reduction, in non-specific binding, an enhancement of binding specificity and an enhancement of binding affinity for a target antigen, compared to non-transformed (wild-type, native) T-cells, are described. The modified TCRs possess an affinity enhancing motif and an affinity weakening motif. Methods of transforming cells and methods of using enriched populations of transformed cells, in the treatment of cancer and infections arid T-cell mediated pathologies are provided. The affinity weakening motif imparts a weakened interaction, of a TCR with major histocompatibility complex proteins, such as HLA proteins in humans.

The device has a porous membrane with top and bottom surfaces and a plurality of pores extending between the top and bottom surfaces. The porous membrane defines a first cell culture region disposed on the top surface and an opposed second cell culture region disposed on the bottom surface. The electrophysiological recording device has a plurality of electrodes positioned on the porous membrane in between a first insulation layer and a second insulation layer. The plurality of electrodes extend to the first cell culture region of the porous membrane. Recording ends of the electrodes measure electrical properties of cells cultured within the first and second cell culture regions, while the contact ends of the electrodes are positioned in electrical communication with data acquisition equipment.
