Publication number: 20250259710Abstract: An image acquisition and analysis system are disclosed. The system enables high throughput, objective analysis of microbial samples over days or weeks. The system may accommodate upwards of twelve 96- or 384-well plates simultaneously (liquid or solid media). The system may acquire and analyze a large number of samples in a short period of time. For example, over 384 samples per minute or 18,432 samples per hour. The system hardware may include a multi-spectral imager (fluorescence and bright field detection), electro-mechanical assemblies, and an optional high-resolution stage. The system may automate image acquisition, image data processing, simplify data storage, and enable automated analysis tools to significantly reduce the manual labor and time associated with such tasks. The system may allow for quick processing and analysis of data into clear phenotypic classes.Type: ApplicationFiled: September 20, 2024Publication date: August 14, 2025Inventors: Matthew Ian WRIGHT, Adrian Christopher SCOTT, Sean Matthew MacLEOD, Megan Lew KUHN

Patent number: 12378696Abstract: This disclosure relates to methods of generating antibody libraries, antibody libraries produced using such methods, and variant antibodies. Presently, methods of improving antibody binding (affinity maturation assays) require the screening of vast libraries of antibody variants (often >1010) to identify a small fraction of variants with improved characteristics. The present invention involves taking the nucleotide sequence of the framework and complementarity determining region of a target antibody and identifying motifs which would be recognised by deamination somatic hypermutation enzymes. A small library of variants is then created which incorporate one or more of these mutations. It was found that a relatively high proportion of the variants have an increased affinity. The technique of the present invention was demonstrated on the trastuzumab and Cathepsin S antibodies, and the variants produced are also claimed.Type: GrantFiled: October 22, 2019Date of Patent: August 5, 2025Assignee: Fusion Antibodies PLCInventors: Richard Buick, Christopher Scott, Darragh MacCann, Leona McGirr, Hugh Morgan, Natasha Campbell, James McClory, Anthony O'Kane

Patent number: 12376370Abstract: A method of fabricating an integrated circuit includes forming a first opening having a first width and a second opening having a second width in a first dielectric layer over a silicon substrate. The openings expose the silicon substrate and the exposed silicon substrate is oxidized to form first and second LOCOS structures having a first thickness. A polysilicon layer is formed over the silicon substrate, so that the polysilicon layer fills the first and second openings. A blanket etch of the polysilicon layer is performed to remove at least a portion of the polysilicon layer over the second LOCOS structure while leaving the first LOCOS structure protected by the polysilicon layer. The silicon substrate under the second LOCOS structure is further oxidized such that the second LOCOS structure has a second thickness greater than the first thickness.Type: GrantFiled: April 11, 2024Date of Patent: July 29, 2025Assignee: Texas Instruments IncorporatedInventors: Abbas Ali, Christopher Scott Whitesell, John Christopher Shriner, Henry Litzmann Edwards
