    Abstract: Disclosed herein are high Tm RNA nanostructures that can be composed of one or more modules or motifs to build RNA nanostructures with or without layers. The RNA nanostructures can have a core domain and three or more double-stranded arms and formulations thereof to conjugate high copy numbers of therapeutics, pH responsive or enzyme cleavable drug cargo. Also described herein is a design strategy for generation of synthetic RNA oligonucleotides that can self assemble into highly thermostable RNA structures. Also described herein are uses of the RNA nanostructures described herein.

    Abstract: A modified immune effector cell may be one in which the functions of a T cell antigen receptor (TCR) and major histocompatibility complexes (MHCI, MHCII) in the modified immune effector cell are inhibited in a T cell. Such a modified immune effector cell may knock out TCR and HLA-A genes expressed by the cell while recognizing surface antigens of tumor cells, so that multiple effects of improving the anti-tumor effect of CAR-T cells, prolonging the survival time of the cells, and reducing the immune rejection response caused by allogeneic cell therapy may be reduced.

    Abstract: An application of a hydrogel in the preparation of products for the treatment of bacterial infection is disclosed. Ferrous ion was used in the antibacterial activity test of Staphylococcus aureus and showed a strong bactericidal effect, which the survival rate of Staphylococcus aureus is less than 0.001%, as well as the survival rate of methicillin-resistant Staphylococcus aureus (MRSA) was less than 0.01%. Ferrous ion hydrogel was used for the treatment of keratitis and skin wound infection. Ferrous ion hydrogel for the treatment of keratitis and skin wound infection, which can significantly reduce the risk of pulmonary MRSA infection, prevented the dissimilation of MRSA to the lung, and alleviated systemic inflammation in the body in a timely and effective manner, revealing the therapeutic potential of ferrous compounds for treatment of Staphylococcus aureus infections, including MRSA.

    Abstract: The present disclosure relates to use of a Clostridium ghonii spore combined with pembrolizumab in cancer treatment. It is found for the first time that the Clostridium ghonii spore combined with pembrolizumab can significantly improve a curative effect of colon cancer and reduce a dose of the pembrolizumab, and thus is efficient and low-toxic. Oncolysis by Clostridium ghonii can affect immunogenicity of a tumor microenvironment (TME) by various ways, converts an immunosuppressive state of the TME into an immune-activated state, adjusts the immunosuppressive TME, and breaks an immune tolerance. An optimal combination of the Clostridium ghonii spore and the pembrolizumab thoroughly removes about 20% of mouse tumor tissues. A benefit range of patients with tumors treated by a PD-1 antibody is expanded. The combination even has an obvious curative effect on patients failed the treatment by the PD-1 antibody.
