    Abstract: This disclosure features methods and compositions for treating diseases of the gastrointestinal tract with an integrin inhibitor. In particular, the disclosure features the topical treatment of sections and subsections of the gastrointestinal tract by integrin inhibitors (like vedolizumab) using an ingestible device that can detect its whereabouts in the Gl tract by using reflectance and which is programmed to release the drugs at a given site or proximal thereto, to treat gastrointestinal inflammatory diseases. The device has hardware storage devices with instructions on board to determine the location and release the formulations. Combination therapies are claimed as well in which the integrin inhibitor is added systemically and another agent (e.g. JAK inhibitor, TNF-alpha inhibitor or IL-12/IL-23 inhibitor) is added topically by means of the ingestible device. The topical administration is said to reduce the side effects associated with systemic treatment.

The innovation can begin by receiving a risk model for validation having multiple sets of data. A first data set is selected from as an input. Outputs are generated for validation. One output can be generating a second set of analysis results using a comparable algorithm to the risk model. Another output can be generating a second set of variables and transformations using a machine learning algorithm and an un-transformed set of the selected variables to assess the set of selected transformations. Another output can be generating a third set of variables using one or more machine learning algorithms and an extended feature set of variables to assess the selected variables. The outputs are compared to the analysis results, coefficients, selected variables, and selected transformations. A report of the comparison is generated.

A user-specific electronic recommendation to address at least one feature identifiable within the pixel data is generated and rendered, on a display screen of a user computing device.
