    Abstract: The present disclosure provides an siRNA capable of inhibiting expression of a complement protein 5(C5) gene, and a pharmaceutical composition and conjugate containing the siRNA. Each nucleotide in the siRNA is an independent modified or unmodified nucleotide. The siRNA comprises a sense strand and an antisense strand. The sense strand comprises a nucleotide sequence I, and the nucleotide sequence I has the same length and no more than three nucleotide differences from a nucleotide sequence shown in SEQ ID NO: 1. The antisense strand comprises a nucleotide sequence II, and the nucleotide sequence II has the same length and no more than three nucleotide differences from a nucleotide sequence shown in SEQ ID NO: 2. The siRNA, the pharmaceutical composition and the conjugate thereof provided by the present disclosure can effectively treat and/or prevent complement-mediated related diseases, such as myasthenia gravis (MG).
