  • Publication number: 20240001497
Abstract: An automatic locking device for assembling a part to a workpiece is provided. The device includes a feeding mechanism, a fixture mechanism, a loading and unloading mechanism, a lifting mechanism, a rotating mechanism, and a locking mechanism. The feeding mechanism feeds the part to the fixture mechanism. The loading and unloading mechanism places the workpiece on the part. The lifting mechanism lifts up the fixture mechanism together with the part and the workpiece. The rotating mechanism rotates the fixture mechanism. The locking mechanism locks the part to the workpiece after the part and the workpiece are rotated by the rotating mechanism. The automatic locking device integrates multiple functions in a single station, reduces the number of the work stations and operatives, lowers the cost, and improves accuracy of the assembly. An assembling apparatus including the automatic locking device is also provided.

  • Publication number: 20230159935
Abstract: The lack of blood-brain barrier (BBB) penetrating ability has hindered the delivery of many therapeutic agents for tauopathy therapeutic treatment. A circular bifunctional aptamer reported here has been able to enhance the in vivo BBB penetration for improved therapy. The circular aptamer includes one transferrin receptor (TfR) aptamer to facilitate TfR-aptamer recognition-induced transcytosis across BBB endothelial cells, and one Tau protein aptamer selected to inhibit Tau phosphorylation and other tauopathy-related pathological events in the brain. This bispecific construct exhibits strong specificity towards Tau and enhanced plasma stability in comparison to linear Tau aptamer. In vivo administration of circular Tau-TfR aptamer results in a rapid uptake into relevant brain regions after crossing the BBB, such as hippocampus and cortex.

  • Patent number: 11530424
Abstract: The clustered regularly interspaced short palindromic repeat (CRISPR) gene editing technique, based on the non-homologous end-joining (NHEJ) repair pathway, can efficiently generate gene knockouts of variably sizes. More precise genome editing, either the insertion or deletion of a desired fragment, can be done by combining the homology-directed-repair (HDR) pathway with CRISPR cleavage. HDR-mediated gene knock-in experiments are inefficient, with no reports of successful gene knock-in with DNA fragments larger than 4 kb. Targeted insertion of large DNA fragments (7.4 and 5.8 kb) into the genomes of mouse embryonic stem cells and zygotes, respectively, using the CRISPR/HDR technique without NHEJ inhibitors was performed and indicate that CRISPR/HDR without NHEJ inhibitors can result in highly efficient gene knock-in, equivalent to CRISPR/HDR with NHEJ inhibitors.

  • Publication number: 20220057409
Abstract: A method, device, and kit are provided for temporal diagnostics and clinical treatment of neuro injury, neuro disease, or neuro repair, particularly including clinical treatment with precision medicines for the same therapeutic targets as a subset of the temporal biomarkers. Through the measurement of biomarkers in a biological sample from a subject, with at least one biomarker from each of the early, intermediate, and late phases of suspected injury, disease, or repair from a subject, a determination of a subject's injury, disease, or repair is provided with greater sensitivity and/or specificity than previously attainable. As many clinical inventions such an anti-inflammatories and clot disruptors are effective only during certain phases injury, disease, or repair, this knowledge can be used to clinical effect in mitigating secondary injuries and/or diseases.

Abstract: The invention relies on detection of specific identified proteins, protein breakdown products, and peptide fragments, to diagnose and evaluate traumatic brain injury, spinal cord injury, and any traumatic injury to the CNS in a subject. These analytes (proteins, protein breakdown products thereof, and peptide fragments thereof) are released from injured tissue into blood and/or cerebrospinal fluid, and can be used to identify the central nervous system cell types (i.e. neuron, astrocyte, oligodendrocyte, and the like) or subcellular structure (e.g., axon, dendrites, presynaptic terminal, post-synaptic terminal, and extracellular matrix) affected, and to determine the diagnosis, location, and severity of the injury. Time course measurements of these analytes measured at different times after an injury or suspected injury also are used as tools for diagnosis and prognosis of central nervous system injury.
