Abstract: The present disclosure provides compositions and methods that employ the compositions for conducting pairwise sequencing and for generating concatemer template molecules for pairwise sequencing. The concatemers can be generated using a rolling circle amplification reaction which is conducted either on-support, or conducted in-solution and then distributed onto a support. The rolling circle amplification reaction generates concatemers containing tandem copies of a sequence of interest and at least one universal adaptor sequence. An increase in the number of tandem copies in a given concatemer increases the number of sites along the concatemer for hybridizing to multiple sequencing primers which serve as multiple initiation sites for polymerase-catalyzed sequencing reactions. When the sequencing reaction employs detectably labeled nucleotides and/or detectably labeled multivalent molecules (e.g.

  • Publication number: 20240190411
Abstract: Methods, systems and devices for monitoring the stress of a rail, in particular, of continuous welded rail (CWR), providing a device that includes radar processing mechanisms and circuitry which is mountable on a rail to be monitored. The radar unit is sealed in a housing and projects a beam to a fixed target that is within a detection range of the radar unit. The radar unit also includes circuitry that allows programming of readings to take place at predetermined time intervals so changes and the extent of changes in conditions of the rail may be determined, and communicated to a remotely situated monitoring device, which may take place through a gateway or other communication network. The devices also may be used to measure temperature and other properties, rail conditions within a tunnel, such as proximity of the rail to walls, as well as ground condition, and bridge monitoring.

  • Publication number: 20240191976
Abstract: A projectile partition that serves as a barrier from ballistics and explosions is disclosed. The projectile partition may serve a traditional role of providing a visual barrier while also providing protection from ballistics and explosions originating from an opposite side of the ballistic screen. The projectile partition may comprise one or more panels or plates that are adapted to repel ballistics and blasts. The projectile partition may further comprise a series of interconnected members that together form a frame which serves to position the one or more panels in a relatively upright position so as to form a barrier against ballistics and explosion blasts.

  • Publication number: 20240193467
Abstract: An approach is provided for upgrading containerized applications in cluster(s) in a container orchestration system. State information about the containerized applications is identified and analyzed. Based on the state information, an upgrade path for an upgrade of the containerized applications is determined. Using an artificial intelligence (AI) based container orchestration platform upgrade engine that employs continuous learning for upgrading data and algorithmic models to upgrade containers, a recommendation is generated that the upgrade be a full upgrade or a partial upgrade. A confirmation of the full upgrade or the partial upgrade is received from a user. Components for the full upgrade or the partial upgrade are created. Using the upgrade path and based on the full upgrade or the partial upgrade, the upgrade is performed.

  • Publication number: 20240191454
Abstract: A manhole cover system for a roadway includes a rotationally fixed outer support securable to an interior of a roadway opening, and an inner sleeve positioned inside of the outer support. The outer support and the inner sleeve share a common central axis. A manhole cover is located at an upper vertical end of the inner sleeve, and a threaded arrangement including a support thread of the outer support is engaged with a complimentary sleeve thread of the inner sleeve, such that rotation of the inner sleeve about the central axis moves the inner sleeve and the manhole cover along the central axis.

  • Publication number: 20240188512
Abstract: A combination rake/refuse collector has a rake portion which traps leaves and other debris, a handle portion which contains a hollow tube, a connector portion which connects the hollow tube to a vacuum unit, and a storage/processing container. The vacuum unit and storage processing container are carried via a shoulder harness with two straps on the back of user. The vacuum unit sucks leaves and other debris collected by the rake portion, through the hollow tube and the connector portion, and directs them to the vacuum storage bag where they are shredded and exhausted into the leaf vacuum bag. The handle portion allows the user to direct the rake portion.

  • Publication number: 20240190278
Abstract: Battery powered quadrotors or drones have a limited operation time. To extend operation time, a powered tether can be used. This tether provides power to the drone allowing it to stay up indefinitely. Most tethered drones are captured to the base station. The tether can reel in and out as the drone moves, but the drone can't go higher or further than the maximum length of the tether. If the tether can be automatically disconnected, the drone could fly off for some remote mission, assuming the drone had an onboard power source such as rechargeable batteries. The present invention relates to a self-powered drone tether that comprises a rechargeable drone in flight which is referred to as the rechargeable drone, a drone that carries a powered tether which is referred to as the tether drone, a coupling mechanism between the rechargeable drone and the tether drone, and a base station with a powered tether and tether deployment system.

  • Publication number: 20240189877
Abstract: Processes and apparatuses for converting PFAS. PFAS are heated and introduced to a reactant which will convert the PFAS. The PFAS may be in a stream that is a PFAS enriched stream formed by desorbing the PFAS from an adsorbent which removed the PFAS from a contaminant stream. An effluent from the PFAS conversion reaction is treated in a treatment zone and a treated effluent may be released to the atmosphere.

  • Publication number: 20240195866
Abstract: A system that runs a hosted spreadsheet application can trigger distributed computing to obtain additional computing support on-demand, when needed to assist with computationally intensive spreadsheet tasks or tasks that take too long to execute. The system will identify spreadsheet cells that each include a respective function that depends on the value of a different cell of the spreadsheet. The spreadsheet server will determine whether a task to be performed on the cells is a computationally heavy task. If the system cannot complete at least a threshold number of the functions before a threshold time period expires, or if the task is otherwise computationally intensive, the system will distribute at least some elements of the task to one or more additional computing devices for processing. The additional computing devices will return their results to a client device and/or an intermediate server.

  • Publication number: 20240188514
Abstract: There is provided a method of manufacturing a growing medium. The method comprises accumulating a particulate material. The particulate material comprises particles having a range of particle sizes. The method further comprises agitating the particulate material. Agitation of the particulate material causes smaller particles of particulate material to sink to the bottom of the growing medium under gravity and to thereby displace larger particles of particulate material to the top of the growing medium. There is also provided a growing medium comprising a particulate material defining a volume of particulate material. The particulate material comprises particles having a range of particle sizes. The concentration of smaller particles of particulate material increases from the bottom of the volume of the particulate material to the top of the volume of the particulate material.
