    Abstract: A non-transitory computer readable medium having computer executable instructions stored thereon is provided. The computer executable instructions include a mission definition file (MDF) that includes mission data that describes alternative routes generated by at least one mission planner for a vehicle. An MDF parser extracts the mission data for the alternative routes from the MDF and maps the mission data into a temporal data structure describing the alternative routes. The temporal data includes a route list to describe the alternative routes for the vehicle and a route segment list to describe an estimated time between each of the waypoints for each segment of the route list. A waypoint list describes a latitude, a longitude, and an altitude for each of the waypoints and a mission plan constructor generates a network topology of a mission plan depicting the alternative routes for the vehicle over time.

    Abstract: Medulloblastoma (MB) is the most common malignant brain tumor of children. To identify the genetic alterations in this tumor type, we searched for copy number alterations using high density microarrays and sequenced all known protein-coding genes and miRNA genes using Sanger sequencing. We found that, on average, each tumor had 11 gene alterations, markedly fewer than in common adult cancers. In addition to alterations in the Hedgehog and Wnt pathways, our analysis led to the discovery of genes not previously known to be altered in MBs. Most notably, inactivating mutations of the histone H3K4 trimethylase genes MLL2 or MLL3 were identified in 16% of MB patients. These results demonstrate key differences between the genetic landscapes of adult and childhood cancers, highlight dysregulation of developmental pathways as an important mechanism underlying MBs, and identify a role for a specific type of histone methylation in human tumorigenesis.

    Abstract: A tire-based system and method for estimating a radially outward tire surface temperature of an identified tire supporting an identified vehicle includes a temperature sensor to measure a tire inner liner temperature. An algorithmic prediction model is empirically trained to correlate inner liner tire temperature to tire radially outward surface temperature for the tire/vehicle combination. The empirically trained algorithmic prediction model receives a steady state temperature-based model inputs including the tire inner liner temperature and an ambient temperature measurement and transient behavior vehicle-based inputs. Based upon the steady state inputs and the transient behavior inputs, a radially outward tire surface temperature estimation is made.

    Abstract: Given the important role of protein kinases in pathways affecting cellular growth and invasion, we have analyzed 340 serine/threonine kinases for genetic mutations in colorectal cancers. Mutations in eight genes were identified, including three members of the phosphatidylinositol-3-kinase (PI3K) pathway; the alterations in the latter genes each occurred in different tumors and did not overlap with mutations in PIK3CA or other non-serine-threonine kinase (STK) members of the PI3K pathway, suggesting that mutations in any of these genes had equivalent tumorigenic effects. These data demonstrate that the PI3K pathway is a major target for mutational activation in colorectal cancers and provide new opportunities for therapeutic intervention.

    Abstract: Analysis of 13,023 genes in 11 breast and 11 colorectal cancers revealed that individual tumors accumulate an average of ˜90 mutant genes but that only a subset of these contribute to the neoplastic process. Using stringent criteria to delineate this subset, we identified 189 genes (average of 11 per tumor) that were mutated at significant frequency. The vast majority of these genes were not known to be genetically altered in tumors and are predicted to affect a wide range of cellular functions, including transcription, adhesion, and invasion. These data define the genetic landscape of two human cancer types, provide new targets for diagnostic and therapeutic intervention and monitoring.

    Abstract: Protein kinases are important signaling molecules involved in tumorigenesis. Mutational analysis of the human tyrosine kinase gene family (98 genes) identified somatic alterations in ˜20% of colorectal cancers, with the majority of mutations occurring in NTRK3, FES, GUCY2F and a previously uncharacterized tyrosine kinase gene called MCCK/MLK4. Most alterations were in conserved residues affecting key regions of the kinase domain. These data represent a paradigm for the unbiased analysis of signal transducing genes in cancer and provide useful targets for therapeutic intervention.
