    Abstract: Efforts to use RNA-cleaving DIMA enzymes (DNAzymes) as gene silencing agents in therapeutic applications have stalled due to their low efficacy in clinical trials. Here the present invention reports a xeno-nucleic acid (XNA) modified version of the classic DNAzyme 10-23 that achieves multiple turnover activity under cellular conditions and resists nuclease digestion. The new reagent overcomes the problem of product inhibition limiting previous 10-23 designs using molecular chemotypes with DNA. FANA, and TNA backbone architectures that balance the effects of enhanced biological stability with RNA hybridization and divalent metal ion coordination. In cultured mammalian cells. X 10-23 facilitates persistent gene silencing by efficiently degrading exogenous and endogenous mRNA transcripts. Together, these results demonstrate that new molecular chemotypes can improve the activity and stability of DNAzymes, and may provide a new route for nucleic acid enzymes to reach the clinic.

A set of signal band sensors is included with a conventional OCM, where each sensor includes a photodetector that is able to detect changes in operation along its associated incoming signal band. Control electronics are used to monitor the states of the photodetectors, with the ability to invoke an OCM scan of the complete wavelength range when a power change condition is flagged.

    Abstract: A photonics integrated circuit chip includes first and second optical waveguides formed on a semiconductor substrate. Light input into the first optical waveguide passes through a first optical isolator and a coupler formed on the substrate. The coupler outputs light from the first optical isolator combined with light from an off-substrate pump light source to an off-substrate optical fiber which outputs the combined light into the second optical waveguide for passage through a second optical isolator formed on the substrate. The substrate may also include third and fourth optical waveguides. Light input into the third optical waveguide passes through a third optical isolator and a second coupler formed on the substrate. The second coupler outputs light from the second optical isolator combined with light from the pump light source to an off-substrate second optical fiber which outputs the combined light into the fourth optical waveguide for passage through a fourth optical isolator formed on the substrate.
