Publication number: 20170145367Abstract: It is not understood what causes or influences pattern formation in cells during the development of an organism. When animal/human cells are cultured in a Petri dish the adherent cells attach to the bottom of the dish, whereas the non-adherent cells float in the growing medium. Currently there are no specialized dishes for culturing non-adherent cells. We now show that non-adherent cells could be induced to form distinct patterns when cultured in an etched plastic dish (Biosimulator). The non-adherent cells showed polarity when cultured in the etched plate. The polarity/pattern formation could be reversed with inhibitors specific for adhesion proteins. The phenomenon of pattern formation by non-adherent cells has wide applications in cell and developmental biology, diagnostics, microbiome research, biofluidics, drug discovery, industrial production of biological products, and also in biotechnology and bioengineering.Type: ApplicationFiled: November 21, 2015Publication date: May 25, 2017Inventor: Sunil Thomas

Publication number: 20170145385Abstract: A method of producing mesenchymal stem cells from induced pluripotent stem cells in which induced pluripotent stem cells are cultured in the presence of a TGF-?inhibitor an in an atmosphere containing from about 7 vol. % to about 8 vol. % C02 for a period of time from about 20 day to about 35 days. The cells then are transfered to a culture dish having a hydrophilic sur face, and the cells are cultured in a medium containing a TGF-? inhibitor for a period of time sufficient to produce mesenchymal stem cells. Such mesenchymal stem cells are more stable and less likely to form tumors, cancers, or teratomas. Also, the induced pluripotent stem cells may be genetically engineered with at least one polynucleotide encoding a therapeutic agent and then are cultured as hereinabove described to provide genetically engineered mesenchymal stem cells that express sustained amounts of a biologically active protein or polypeptide.Type: ApplicationFiled: July 8, 2015Publication date: May 25, 2017Inventors: Darwin J. Publication number: 20170145386Abstract: A composition comprising a plurality of cell aggregates for use in the production of engineered organotypic tissue by organ printing. A method of making a plurality of cell aggregates comprises centrifuging a cell suspension to form a pellet, extruding the pellet through an orifice, and cutting the extruded pellet into pieces. Apparatus for making cell aggregates comprises an extrusion system and a cutting system. In a method of organ printing, a plurality of cell aggregates are embedded in a polymeric or gel matrix and allowed to fuse to form a desired three-dimensional tissue structure. An intermediate product comprises at least one layer of matrix and a plurality of cell aggregates embedded therein in a predetermined pattern. Modeling methods predict the structural evolution of fusing cell aggregates for combinations of cell type, matrix, and embedding patterns to enable selection of organ printing processes parameters for use in producing an engineered tissue having a desired three-dimensional structure.Type: ApplicationFiled: February 3, 2017Publication date: May 25, 2017Inventors: Gabor Forgacs, Karoly Jakab, Adrian Neagu, Vladimir Mironov
