Abstract: The invention relates to methods for producing lipids such as fatty acid products in recombinant host cells engineered to express a non-native gene encoding a dehydrogenase. The recombinant microorganisms are able to proliferate at a higher rate as compared with microorganisms that do not express a non-native dehydrogenase gene, and cultures of microorganisms that are engineered for lipid production and that express a non-native dehydrogenase produce more lipid than cultures of control microorganisms that do not include a non-native dehydrogenase gene.

Abstract: This disclosure pertains to the functional localization of heterologous integral plasma membrane proteins (HIPMPs) lacking cleavable signal sequences into the plasma membrane (PM) of cyanobacterial hosts, e.g., JCC138 (Synechococcus sp. PCC 7002) or an engineered derivative thereof. More specifically, the disclosure provides chimeric integral plasma membrane proteins comprising pseudo leader sequences (PLSs) that promote increased hydrocarbon (e.g., alkane) export capabilities when expressed in a photosynthetic organism, e.g., a cyanobacterium.

Abstract: The present disclosure identifies pathways and mechanisms to confer improved industrial fitness on engineered organisms. It also discloses engineered organisms having improved industrial fitness. Synthetic biologic engineering modules are disclosed that provide for light capture, carbon dioxide fixation, NADH production, NADPH production, thermotolerance, pH tolerance, flue gas tolerance, salt tolerance, nutrient independence and near infrared absorbance. The disclosed engineered organisms can include one or more of these modules. Also provided are methods of using the engineered organism to produce carbon-based products of interest, biomass or pharmaceutical agents.

Abstract: The invention provides eukaryotic unicellular algae engineered to express a nucleosome alteration protein fused to a protein with affinity to the DNA binding site acting in coordination. An example is a LexA-p300 fusion protein, where the p300 is derived from Chlamydomonas. The LexA binding domain guides the p300 to the binding site and the p300 loosens the nucleosome structure by acetylating histones within proximity of the transgene, thus remodeling the local chromatin structure to allow for high-level expression.

Abstract: A transformed diatom includes a nucleic acid sequence operatively linked to a promoter, wherein the nucleic acid sequence encodes an amino acid sequence including (i) an heterologous signal peptide and (ii) a polypeptide, the heterologous signal peptide leading to the secretion of the polypeptide in the extracellular medium of the transformed diatom; a method for producing a polypeptide which is secreted in the extracellular medium, the method including the steps of (i) culturing a transformed diatom, (ii) harvesting the extracellular medium of the culture and (iii) purifying the secreted polypeptide in the extracellular medium; and use of the transformed diatom for the secretion of a polypeptide in the extracellular medium.
