    Abstract: Drug-loaded microbead compositions include microbeads of a water-swellable polymer material and a complex of a carrier and a therapeutic agent chemically bonded to the carrier. The complex is embedded in the polymer material. Methods for preparing the drug-loaded microbead compositions and embolization compositions include loading a therapeutic agent into a water-swellable polymer material to form microbeads, then removing water from the microbeads. Additional drug-loaded microbead compositions include microbeads of a biodegradable material, vesicular agents, a first therapeutic agent associated with the vesicular agents, and a second therapeutic agent different from the first therapeutic agent. The vesicular agents include a lipid bilayer surrounding a vesicular core. The coating layer comprises a hydrophobic therapeutic agent and at least one additive. The modified exterior surface may affect the release kinetics of the drug from the device, the crystallinity of the drug layer, the surface morphology of the coating and particle shape, or the particle size of drug of a therapeutic layer in the coating layer. For example, the effects caused by the modified exterior surface may increase the retention time and amount of therapeutic agent in tissue.

    Abstract: A novel prophylactic therapy for cancer, as well as therapy against DNA damage, inflammation and immunosuppression involves the ?-blocker carvedilol, which is a racemic mixture consisting of two enantiomers, S- and R-carvedilol, in 1:1 ratio. S-carvedilol is a ?-blocker, with a highly potent antagonizing activity against the ?-adrenergic receptors, which is the main mechanism for the drug's pharmacological activity in treatment of high blood pressure and heart failure. Carvedilol—the racemic mixture—prevents ultraviolet radiation induced skin cancer by attenuating DNA damage, reducing inflammation and reversing immunosuppression. The non-?-blocking enantiomer R-carvedilol exhibits the same cancer preventive efficacy as the racemic carvedilol, without disturbing the cardiovascular system. Both carvedilol and R-carvedilol prevent chemical carcinogen-induced lung cancer development and lung inflammation.
