  • Patent number: 6040430
Abstract: An invention is described that permits the regulation of recombination in cells, organisms or appropriate cell-free systems. The invention involves creating fusion proteins between recombinase proteins, or components of recombinase systems, and ligand binding domains derived from nuclear receptors. The fusion proteins show little recombinase activity in the absence of the ligand that binds to the ligand binding domain. Upon binding of the ligand, recombinase activity is induced. The invention provides a practical means to regulate recombination in cells and organisms and, by linking ligand binding to recombination, provides a simple means whereby ligand binding can be measured as recombination achieved.

  • Patent number: 5053326
Abstract: In order to carry out hybridization reactions by forming a complex between a polynucleotide and a detectable substance and using the complex thus formed for hybridization reactions, a single strand polynucleotide is reacted with a positively charged nucleic acid binding detectable protein or polyamine in solution and the resultant complex is covalently bound with a cross-linking agent and the polynucleotide compound obtained is used as a marker in hybridization experiments in the elucidation of complementary sequences in foreign polynucleotides. The protein may be an enzyme such as horseradish peroxidase modified by a polyamine e.g. a polyethylene imine to provide a positively charged region. Assay sensitivity may be enhanced by performing the hybridization in the presence of about 6% (w/v) of a polyethylene glycol.

  • Patent number: 4533307
Abstract: An apparatus for preparing thin layers, such as thin gel layers useful in electrophoresis, wherein a liquid layer material is introduced to the space defined between two parallel mold surfaces on first and second mold elements. Initially, the mold surfaces overlap each other to a small extent, and liquid is introduced to the space between the surfaces adjacent the leading edge of the top mold element. The mold elements are then shifted with respect to each other while maintaining their parallel relationship, and additional liquid layer-forming material is introduced to the space during the shift. In a preferred embodiment, a third mold element is situated adjacent to and spaced a small distance from the bottom mold elements, and liquid material is introduced to the space between the bottom mold elements during shifting of the top mold.

  • Patent number: 4532205
Abstract: A method and a device are proposed for the injection of very small amounts of samples into cells (74) with the aid of a microcapillary (10). During the injection, a constant injection pressure of about 0.12 bar, provided by a pressurized cylinder (46) is applied to the microcapillary (10), which is reduced after the completion of the injection to a holding pressure of 0.02 bar until the next cell (64) is to be injected. A high-pressure (p.sub.1) supplied from another pressurized cylinder (44) serves for blowing out the microcapillary tip (12). A three-way valve (42) permits simple and rapid switching between the various pressures (p.sub.1, p.sub.2 and p.sub.3). Reproducible amounts of sample in the range from 10.sup.-11 ml to 10.sup.-13 ml can be injected reproducibly, simply and rapidly.
