Abstract: Described herein is a method of preparing an unbiased library of phage variants, comprising (a) preparing a population of “acceptor phage”; (b) removing an endogenous target gene and inserting gene variants into the acceptor phage genomes; (c) enriching the recombined phages; and (d) expressing the library for selection. The acceptor phage is a lytic phage comprising a synthetic genome wherein the target gene of interest is flanked by recombinase sites. The acceptor phage infects a first host bacteria expressing a recombination plasmid facilitating recombination. The phages then infect a second host bacteria expressing a counterselection system that accumulates recombined phage variants and selecting against non-recombined phages. The accumulated phage variants infect a third host bacteria. The phage library may then be sequenced and characterized.

Abstract: The present invention relates to a novel biomarker for diagnosing cancer, and a use thereof. STC1, a novel biomarker for diagnosing or predicting the prognosis of cancer according to the present invention, was found to be related to poor prognosis of cancer patients according to the expression level, and was found to be overexpressed in various cancer cell lines. In addition, STC1 was found to be a biomarker related to the proliferation, invasion, and migration (metastasis) of cancer cells. In addition, it was found that STC1 is detected in the serum or urine of bladder cancer patients, and can be effectively used for diagnosing and predicting the prognosis of bladder cancer by identifying differences in expression according to the patient's clinical stage.

Abstract: The present disclosure relates to an upstream of mTORC2 (UT2) gene-deficient mouse model and a method of screening therapeutic agents for myeloid leukemia using the same, wherein it was found that the onset of myeloid leukemia and proliferation of myeloid leukemia cells were more facilitated in UT2 gene-deficient mice and UT2 genetic scissor (Cripsr/Cas9)-deficient myeloid leukemia cell lines. In addition, it was found that the survival rate of patients with low expression of UT2 was low in myeloid leukemia patients, while expression of UT2 was low in cells of actual myeloid leukemia patients.
